A Comprehensive Study On Nifedipine

Objective:To evaluate the efficacy.safety, economic, Social Property and drug information of nifedipine and amlodipine in hypertensive by using meta-analysis to provide evidence for the clinical use of medicines.Methods:Retrieved from CNKI, Wanfang Database and VIP Information Resource System,MEDLINE(EBSCO),EMBASE (Ovid):1980 to 2009 week 46,Pubmed,Cochrane library, evaluate the quality of the literature search,meta-analysis was conducted using Review Manager 5.0.25 software.Results: ① The paper met the inclusion criteria were 74 of the nifedipine and amlodipine effectiveness evaluation, Meta-analysis showed:The total nifedipine and amlodipine treatment of hypertension statistically significant difference in efficiency: OR=0.47,95%CI[0.40,0.56],(P=0.00001)<0.1 The OR<1, the forest mapdiamond fell on the left side of ineffective line,showing that the effectiveness of nifedipine was lower than amlodipine group. heterogeneity with (P=0.02)<0.1 and I2=27%, prompted the research of homogeneity.using fixed effect model to calculate the combined statistics; Funnel plot efficiency of the data was overall symmetrical, but something was missing in the bottom left corner, indicating that there was still publication bias. ② The paper met the inclusion criteria were 60 of the nifedipine and amlodipine safety evaluation, Meta-analysis showed: The total nifedipine and amlodipine treatment of hypertension statistically significant difference in safety:OR=3.36,95%CI[2.75,4.11],(P=0.00001)<0.1. The OR>1, the forest map diamond fell on the right side of ineffective line.showing that the adverse reactions of nifedipine was greater than amlodipine group, heterogeneity with ((P=0.001))<0.1 and I2=40%, prompted the research of homogeneity.using fixed effect model to calculate the combined statistics; Funnel plot safety of the data was overall symmetrical, but something was missing in the bottom left corner, indicating that there was still publication bias.③Pharmacoeconomic analysis showed that amlodipine was smaller in cost—effectiveness than nifedipine.Conclusion:The available clinical evidence suggests that nifedipine of the total efficiency is less than the total efficiency of amlodipine; nifedipine is greater than the incidence of adverse reactions amlodipine; nifedipine GITS cost’·effectiveness is greater than amlodipine,amlodipine,compared between the two is the best treatment.
