Cold and heat are the core issues of Traditional Chinese Medicine (TCM). No matter the exploration of yin-yang theory, the research on six-meridian syndrome, or the study of the Chinese herb properties, are all involved cold and heat issues directly or indirectly. Four properties of Chinese herbs reflect their function tendency of Yin-Yang exuberance and debilitation, and cold and heat changes in human body. Due to different historical era, practical experience, level of understanding, physicians in successive dynasties tend to have different understandings of cold and heat property. Unclear or unknown property can not meet the demands for clinical practice, but either influences the accuracy of clinical prescription to some extent. As a result, combined with modern technology, the research on objectivity of cold and heat degree of Chinese herbs is extremely necessary.It was found in previous studies that the body temperature changes after Chinese herb’s reaction is an indicator to measure the degree of cold and heat property of Chinese herb, which preliminarily proves that the body temperature changes after drug’s reaction is an indicator to measure the degree of cold and heat property of Chinese herb. Microcalorimetry can online and sensitively reflect the overall heat production and specific details of organism after administration of drugs in real time, but there are still differences in measurement methods system and technical indicators. This study explores and preliminarily establishes a research method of cold and heat property of Chinese herbs based on microcalorimetry by observing the influence of bacterial cultivation system, bacterial inoculum size and intervention concentration of Chinese herbs on colibacillus’ exothermic curve.Objectives:1.To preliminarily create the technology method of detecting cold and hot property of Chinese herbs with visualization, quantitative and repeatable technology based on microcalorimetry;2.To conduct otherness study of cold and heat degree of representative Chinese herbs for six-meridian differentiation and treatment in Treatise on Cold-Induced Diseases including radix aconiti carmichaeli, rhizoma zingiberis, fructus evodiae, poria, rheum officinale, scutellaria baicalensis, rhizoma coptidis by the application of microcalorimetry.Methods:1.Theoretical StudyBased on the essence of six meridians and holism of pathological changes, quantification thoughts of cold and heat syndrome differentiation in Treatise on Cold-Induced Diseases were summarized according to six-meridian syndromes. Through relative research on typical classics on Chinese herbal medicine, four properties of 52 kinds of vegetable Chinese herbs in Treatise on Cold-Induced Diseases were selected And cold and heat property changes of representative Chinese herbs for six-meridian differentiation and treatment in Treatise on Cold-Induced Diseases including radix aconiti carmichaeli, rhizoma zingiberis, fructus evodiae, poria, rheum officinale, scutellaria baicalensis, rhizoma coptidis were analyzed.2.Experimental researchBased on microcalorimetry theory, systematic research was conducted from three aspects of bacterial cultivation system, bacterial inoculum size and intervention concentration of Chinese herbs to observe the influence of different bacterial cultivation system, bacterial inoculum size and intervention concentration of Chinese herbs on colibacillus’growth and exothermic curve. Furthermore, the established microcalorimetry method was applied to observe the influence of decoction of radix aconiti carmichaeli, rhizoma zingiberis, fructus evodiae, poria, rheum officinale, scutellaria baicalensis, rhizoma coptidis on colibacillus’ growth and exothermic curve.Results:1.Theoretical researchSix-meridian syndrome differentiation theoretical system was firstly created in Treatise on Cold-Induced Diseases, and cold and heat syndrome differentiation was going through the whole book as the main evidence to differ the six-meridian syndromes. Zhang Zhongjing did quantitative syndrome differentiation to six-meridian syndromes from various aspects of course of disease, symptoms, treatment and prescriptions, which was the specific embodiment of thoughts of syndrome differentiation and treatment varying with each individual and syndrome. Understanding Zhang Zhongjing’s method of using quantitative thoughts to analyze disease’s dynamic changes and comprehending nuanced, changeable and regular syndrome differentiation thoughts with strong systematicness and holistic consistency are significant to learn, understand and practice Zhang Zhongj ing’s academic thougths, which can make syndrome differentiation and treatment more accurate, meticulous and meticulous.Classic books about Chinese herbs in all ages had quantitative descriptions of Chinese herb’s four properties with great cold, cold, slight cold, natural, warm, slightly warm, heat, and great heat, but the understanding varied. For example, in 52 kinds of vegetable Chinese herbs in Treatise on Cold-Induced Diseases, there were different records of properties of nearly 50%(25 kinds) of these Chinese herbs. Some Chinese herbs had different cold and heat or warn and heat degree, while others had great difference in cold and heat property. So objectivity research on cold and heat was in urgent need. Cold and heat property of 7 Chinese herbs of radix aconiti carmichaeli, rhizoma zingiberis, fructus evodiae, poria, rheum officinale, scutellaria baicalensis, rhizoma coptidis were textually criticized, and it was proved that there were not any controversies of cold and heat property. These Chinese herbs were chosen as representative Chinese herbs to interfere colibacillus’ growth curve, and empirical study by microcalorimetry was conducted.2.Experiment researchColibacillus’ growth and exothermic cures in four cultivation systems with 5ml,10ml, 15ml and 20ml, eight inoculum sizes with 5×107,2.5×107,5×106,2.5×106,5×05,2.5×105, 5×104,2.5×104 and seven decoction concentrations with 2g/ml, Ig/ml,0.5g/ml,0.25g/ml, 0.125g/ml,0.0625g/ml,0.03125g/ml were compared and analyzed, and microcalorimetry research method of Chinese herb cold and heat property with 5ml cultivation system, 2.5x105cfu/ml bacterial inoculum size and 0.5g/ml Chinese herb interference concentration. Based on this, it was proved that scutellaria baicalensis, rhizoma coptidis with cold property could delay the appearance time or decrease the peak power of colibacillus’ growth curve. Radix aconiti carmichaeli, rhizoma zingiberis, fructus evodiae with heat property had the tendency of improving bacterial growth.Conclusion:The study proved that all of reaction system, carrier bacterial concentration, and Chinese herb interference concentration could have influence on bacterial exothermic cures. The microcalorimetry method that preliminarily created in this study could be applied into the objectivity research on cold and heat property of Chinese herbs. The appearance time, peak power and overall heat release of colibacillus were major technical parameters to reflect cold and heat degree of Chinese herbs.
