| Objective:To observe the effects of oxaliplatin on cell growth inhibition and apoptosis induction in human bokitt lymphoma carcinoma cell line Raji, for offering accordance with the theory of the clinical application.Methods:Raji cells were incubated with oxaliplatin at various concentration and time (0,6.25,12.5,25,50,100μg/ml and 24,36,48h), cell growth inhibition was assessed by CCK-8 methods, cell-cycle kinetics and apoptosis were analyzed by fluorenscence microscopy and flow cytometry. using the method of immunohistochemisty to detect the protein effect of ki-67and bax. (with oxaliplatin at various concentration and time (0,6.25,12.5,25,50,100μg/ml and 48h).Results:The growth of Raji cells was significantly inhibited by oxaliplatin in a dose- dependent and time-dependent fashion. The inhibition of cell growth was (94.73±1.40)% after incubation with 100μg/ml oxaliplatin for 48 hours.oxaliplatin could cause Raji cells cycle to change. With the increase of drug concentration, The earl-stage and late-stage apoptosis rate of Raji cells were (8.25±1.79)% and (14.61±2.18)% after incubation with 100μg/ml Oxaliplatin for 48 hours respectively. ki-67 protein concentration to descend with oxaliplatin congcentration raise gradually. And Bax protein concentration to raise with oxaliplatin congcentration raise gradually.Conclusions:Oxaliplatin suppresses the growth of Raji cells in vitro by causing cell-cycle arrest and cell apoptosis.Oxaliplatin can affect the protein expression of Ki-67 and Bax. Oxaliplatin promotes the cell apoptosis effect of the burkkit-Raji cell could be concerned with up-regulation of the Bax protein. |
