| Objective To study the effect of mangiferin on the growth inhibition and invasive ability of human lymphoma Raji cells.Methods Raji cells were given different concentrations of mangiferin. The inhibition ratio of the cells were measured by MTT assay and invasive ability were measured by Minicell chamber assay.Results The results of MTT assay showed that the cell growth inhibited rate are 48.1%,51.5%,65.3%,68.9%,81.5%(P<0.01)in dose-time dependent manner after treated by (25μmol/L,50μmol/L,100μmol/L,150μmol/L, 200μmol/L) mangiferin for 72 hours, and the cell growth inhibited rate are 44.9%,71.2%,81.5%(P<0.01) respectively with 200μmol/L mangiferin treatment for 24h, 48h, 72h. he results of Chamber experiment showed that the number of cells are 172.1±4.5,167.2±3.4,155.5±4.5,146.8±2.8,121.1±3.6 (P<0.01)under micro-membrane decreased in raji cells, in dose dependent manner after treated by 25-200μmol/L mangiferin for 24 hours.Conclusion Mangiferin can inhibit obviously the growth and invasive ability of human leukemic Raji cells. Objective: To study the effect of mangiferin on the expression of the related gene tiam1 on lymphoma Raji cells and its possible mechanism.Methods: Raji cells were given different concentrations of mangiferin. Western bolt and RT-PCR were used to detect the expression of the mRNA and protein of TIAM1 gene..Result: In dose-time dependent manner after treated by 50μmol/L,100μmol/L,200μmol/L, Mangiferin efficiently suppressed the expression of taim1 in raji cells for 24h, the expression of tiam1 are 0.311±0.05,0.135±0.04,0.094±0.02(P<0.05) in dose dependent manner. The expression of tiam1 porten in 200μmol/L group was significantly down-regulated than the other two groups (P<0.05).Conclusion: Mangiferin can inhibit the expression of tiam1 mRNA and also can inhibit the expression of porten of tiam1.. |
