| ObjectiveNasopharyngeal carcinoma is a common malignant tumor in head and neck,invasion and metastasis are responsible for the high rate of mortal.The aim of this research is to find more effectively chemotherapeutic agents and treatment regimens, we studied the effects of Oxaliplatin on cell growth inhibition and apoptosis induction in human nasopharyngeal carcinoma line CNE-2.MethodsCNE-2 cells were incubated with Oxaliplatin at various concentration and time, cell growth inhibition was assessed by Sulphorhodamine assay, cell-cycle kinetics and apoptosis were analyzed by fluorenscence microscopy and flow cytometry.ResultsThe growth of CNE-2 cells was significantly inhibited by Oxaliplatin in a dose-dependent and time-dependent fashion. The inhibition of cell growth was (97.46±1.76)% after incubation with 80μg/ml Oxaliplatin for 48 hours. Oxaliplatin could cause CNE-2 cells cycle to change. With the increase of drug concentration, G2/M phase cell percentage increased. The early-stage and late-stage apoptosis rate of CNE-2 cells were (4.48±1.23)%and (9.48±2.30)%after incubation with 80μg/ml Oxaliplatin for 48 hours respectively.ConclusionsOxaliplatin suppresses the growth of CNE-2 cells in vitro by causing cell-cycle arrest and cell apoptosis . |
