Objective: The impacts of Cortex Eucommiae on biochemical markers, bone microstructure and ultrastructure of femurs and lumbar vertebrae in ovariectomized rats with experimental osteoporosis are observed and the role of primary osteoporosis prevention and treatment Cortex of Eucommiae is further discussed.Methods: In this study, 80 October-old female SD (Sprague-Dawley) rats were, in a completely randomized way, divided into four groups: Group sham (n=20), Group OVX(n=20), Group OVX+Alfacalcidol (n=20), GroupOVX+E Cortex Eucommiae (n=20). castration Eucommia gavage group 20. Animal models of osteoporosis were reproduced in Group OVX, Group OVX+Alfacalcidol, and GroupOVX+E Cortex Eucommiae. After oral administration, comparison of serum estradiol (E2) and osteocalcin (BGP) levels were respectively observed, and microscopic and ultrastructural changes in femurs and lumbar vertebrae of ovariectomized rats were observed respectively in the light and electron microscopyResults: 1, E2 expression: Serum E2 in Group OVX, Group OVX+Alfacalcidol, GroupOVX+E Cortex Eucommiae was significantly lower than that in Group sham (P<0.01). Serum E2 in Group OVX+Alfacalcidol, GroupOVX+E Cortex Eucommiae was higher than that in Group OVX.(P<0.05).; 3 BGP expression: serum BGP in in Group OVX, Group OVX+Alfacalcidol , GroupOVX+E Cortex Eucommiae was significantly higher than that in Group sham (P<0.01). Serum BGP in GroupOVX+E Cortex Eucommiae was lower than that in Group OVX+Alfacalcidol (P<0.05).2.The microscope ovariectomized rats in femurs and lumbar vertebrae contain a lot of fat, and trabecular bone density decreased significantly, while fat particles in the sham group, positive control group and experimental group Cortex Eucommiae were rare,and bone trabeculae were more dense. In electron microscope, osteoclasts in femurs and lumbar vertebrae of ovariectomized rat were significantly developed , manifested in the more advanced rough endoplasmic reticulum (RER), lysosomes and mitochondria and other membrane system, larger nuclei, often with a larger proportion of chromatin, and some relatively shrinking osteoblasts, underdeveloped endomembrane system, and reduced cytoplasmic volume. In the castrated rats, part of apoptotic cells, and intracellular lipid droplets in some cells were also observed.Compared with the castration group, in positive control group and experimental group Cortex Eucommiae, relatively underdeveloped endomembrane system, and more lysosome were observed in osteoclasts. But bone lining cells system (except the lysosome) were relatively well-developed in osteoblasts ,and intracellular lipid droplets and apoptotic cells, were also not or seldom found in. osteoblasts.Conclusion: 1, Cortex Eucommiae, by inhibiting osteoclast activity and promoting the vitality of osteoblast, can improve osteoporosis and promote bone formation. 2, Cortex Eucommiae has a significant role in the regulation of the activation of adipocyte. 3 The intervention on bone cells of Cortex Eucommiae is related to inhibition of cell apoptosis.
