Eucommiae cortex is the dried cortex of Eucommia ulmoides Oliv.. It is a commonly used tonic traditional chinese medicine,which is sweet in flavor, warm in nature, and attributive to liver and kidney channel.It can nourish the liver and kidney, strengthen bones and muscles, calm fetus.It was used to treat weak of liver and kidney, pain of the loin and knee, muscles weakness, dizziness, leakage of blood in pregnancy, restless fetus and so on.The doctors in past dynasties used Eucommiae cortex for invigorating the kidney and strengthening the bones,and the modern study of pharmacology suggest that Eucommiae cortex can assuredly cure osteoporosis. For the effective Components on treatment of osteoporosis of Eucommiae cortex are unclear, this thesis screened effective fraction and effective Components on treatment of osteoporosis from Eucommiae cortex. And we studied the method of determination of the effective compounds and make the fingerprint of Eucommiae cortex.The first chapter of this thesis screened the effective fraction on treatment of osteoporosis of Eucommiae cortex by the osteoporotic model of rats caused by retinoic acid and the yang-deficiency model of mices caused by hydrocortisone. The experiment results indicated that n-butanol portion could increase bone density, reduce ALP activity of osteoporotic model of rats and enhance the index of suprarenal gland, thymus gland, prolong the time of loaded-swimming of the yang-deficiency model of mices.Six compounds were isolated from n-butanol portion by chromatography and their structures were elucidated by spectral method as:geniposide acid, aucubin, chlorogenic acid, geniposide, glucose, sucrose.using osteoblast and osteoclast experiments in vitro to detect for activity of five compounds of n-butanol portion:aucubin, geniposide, geniposide acid, chlorogenic acid and pinoresinol diglucoside. The results show that aucubin, geniposide, geniposide acid and pinoresinol diglucoside were the effective components on treatment of osteoporosis.The second chapter of this thesis established a HPLC method to determine the contents of geniposidic acid、chlorogenic acid and pinoresinol diglucoside as index components and detected11batch Eucommiae cortex.The quantity of geniposidic acid、chlorogenic acid and pinoresinol diglucoside in Eucommiae cortex were0.039%-0.124%,0.013%-0.199%.0.106%-0.294%. HPLC finger-print was setted up by11batch Eucommiae cortex from different places which contained the12common peaks.The peak of geniposide, geniposide acid, chlorogenic acid, pinoresinol diglucoside was confirmed.The similitude degree of11batch Eucommiae cortex is between0.82and0.96.The HPLC fingerprint and simultane determination of geniposidic acid、chlorogenic acid、pinoresinol diglucoside were established which provide the scientific basis for the quality control of Eucommiae cortex.
