Objective:To investigate the expression of RUNX3 and P21WAF1/CIP1protein in human cervical squamous cell cancer and cervical intraepithelial neoplasia, to explore the role of RUNX3 and P21WAF1/CIP1 in the tumorigenesis and progression of human cervical squamous cell cancer .speculated the existence of a mechanism in the regulation of cell proliferation,which can provide a new reference to cervical cancer gene therapy .Methods:The expression of RUNX3 and P21WAF1/CIP1 in 20 cases of normal cervix tissue, 45 cases of Cervical Intraepithelial Neoplasia (CIN) and 45 cases of cervical squamous cell cancer tissues were detected by immunohistochemical EliVision two-stages method. And then analyzed statistically.Results:1.RUNX3 protein was detected in normal cervix tissue, cervical Intraepithelial Neoplasia and human cervical squamous Cell carcinoma. The rate of RUNX3 protein expression in cervical intraepithelial neoplasia was considerably higher than that in normal tissuse (P<0.01). The positive expression rate for RUNX3 protein in human cervical squamous cell carcinoma was so much higher than that in cervical intraepithelial neoplasia (P<0.05). the expression of RUNX3 was related to the histopathologic grade, depth of stromal invasion and lymph node metastasis.2.P21WAF1/CIP1 protein was detected in normal cervix tissue, cervical Intraepithelial Neoplasia and human cervical squamous Cell carcinoma. The rate of P21WAF1/CIP1 protein expression in cervical intraepithelial neoplasia was considerably higher than that in normal tissuse (P<0.01). The positive expression rate for P21WAF1/CIP1 protein in human cervical squamous cell carcinoma was so much higher than that in cervical intraepithelial neoplasia (P<0.05). the expression of P21WAF1/CIP1 was related to the lymph node metastasis, independent of other clinicopathological features, Clinical stage, histological grade, tumor invasion and so on.3.There was a positive correlation between RUNX3 and P21WAF1/CIP1 protein in cervical squamous cell cancer.4.High expression of RUNX3 notice Poor prognosis,the 3-year survival rate in low-expression group was higher than the high expression group (P<0.05).Conclusions:The expression RUNX3 and P21WAF1/CIP1 protein are increasing , Their abnormal expression maybe associated with malignant biologial behavior. RUNX3 and P21WAF1/CIP1 expression in cervical squamous cell cancer was positively correlated with each other, there may be interactions between the two. Furthermore, the expression of RUNX3 may provide useful prognosis implication and be an useful prognosis indicator for cervical squamous cell cancer.
