SUBJECTTo compare the different behavior and blood pressure of nomal group, model control group, model group,Chinese crude drug group and western medicine group during whole range,and to detect the expression difference of Tpxâ…¡,Hsp27 and ANXA1 mRNA among these rats.To approach the mechanism of degrading blood pressure and improving hyperactivity of liver-YANG in gene level,and to provide a theoretical basis for the possible targets of calming the liver and suppressing yang drugs.METHODS1. With 10 normal SD rats for the nomal group,40 spontaneously hypertensive rats(SHR) were randomly divided into a model control group(10),a model group(10),a Chinese crude drug group(10) and a western medicine group(10).SHR were made to hyperactivity of liver-YANG model by drenching with prepared Aconiti Praeparatae Decoction,and then were treated by Chinese crude drug or western medicine drugs.2. To Monitor the changes of behavior and blood pressure of rats during being made model and drug treatment,then take their hypothalamic and adrenal gland unified out of at the seventh weekend.RT-PCR was performed to detect the expression of Tpxâ…¡,Hsp27 and ANXA1 mRNA in the hypothalamus,and then made the statistical analysis.RESULTS1. The reproduction and treatment of animal model:After intragastric administration of Aconiti Praeparatae Decoction,the irritability degree of SHR were levelâ…¢mostly,and most rats had a conjunctival congestion,while the drinking water and contractive pressure step up obviously.After the rats treated by Chinese crude drugs or western medicine(the seventh weekend),the conjunctival congestion and irritability degree were all improved,and the drinking water and contractive pressure were less than pretherapy and homochronous model group,but there was no significant difference between the Chinese crude drug group and western medicine group(P>0.05).2. The comparation of expression of three mRNA in adrenal glands:The expression of Tpxâ…¡and ANXA1 mRNA in nomal group were lower than model control group(P<0.05),while the expression of Hsp27 mRNA was no statistical significance with nomal group(P> 0.05).The expression of model group was the highest than the others(P <0.01).The expression of Tpxâ…¡and ANXA1 mRNA were lower then model control group(P<0.05),while the expression of Hsp27 of two groups mean nothing to it,but the expression of three mRNA were stile higher than nomal group(P< 0.05).There were not statistically significant about Hsp27 and ANXA1 comparison in Chinese crude drug group and western medicine group(P> 0.05),but Tpxâ…¡mRNA in Chinese crude drug group was lower than western medicine group(P< 0.05).3. The comparation of expression of three mRNA in hypothalamuses:The expression of Tpxâ…¡and ANXA1 mRNA in nomal group were lower than model control group(P< 0.05),while the expression of Hsp27 mRNA was no statistical significance with nomal group(P>0.05). The expression of model group were higher than the model control group in different degree(P<0.01).The expression of all mRNA in Chinese crude drug group and western medicine group were fewer than model group,but higher than nomal group(P<0.05).There was no statistical significance about the comparison of expression of three mRNA in Chinese crude drug group and western medicine group(P>0.05).CONCLUSION1. Calming the liver and suppressing yang drugs can improve the behavior and blood pressure of the SHR with hyperactivity of liver-YANG.2. The blood pressure of SHR with hyperactivity of liver-YANG was reduced with symptoms ameliorating after treated by Chinese crude drugs,which might be related to the adjustment of Tpxâ…¡,Hsp27 and ANXA1 mRNA in hypothalamuses and adrenal glands. |
