| [OBJECTIVE] To investigate the relationship of Helicobacter pylori Infection, IL-10 polymorphism and susceptibility to gastric cancer in high incidence area of Hexi area of Gansu province.[METHODS] All investigated objects were divided into three groups:151 gastric cancer patients,148 precancerous lesion patients and 153 control subjects were enrolled. Patient clinical data were collected, including age, gender, epidemiological data, family history of gastric cancer, endoscopy or after the diagnosis of pathology results.The Interleukin-10-1082 promotr polymorphism was detected by SNaPshot; and Immunoblotting was performed to test H.pylori. The results were analyzed by SPSS15.0[RESULTS]:①The distribution frequency of AA,AG/GG two genotypes were75.8% ,24.2% respectively in normal control.②The frequency of AG+GG genotype was statistically higher in the gastric precancerous lesion (GPL) group compared to the control group (P=0.018), moreover, individuals with the IL-10AG+GG genotype increased 1.801 fold risk for gastric precancerous lesion.③The frequency of AG+GG genotype was conspicuous higher in the gastric cancer (GC) group compared to the control group (P=0.010), moreover, individuals with the IL-10AG+GG genotype increased 2.244 fold risk for gastric cancer.④Participations exposed in both enviromental factor (H. pylori infection) and gene mutation factor (carrying IL-10-1082 AG/GG genotypes), the risk of developing gastric precancerous lesion and gastric cancer was 4.767,7.55 fold of those exposed in neither enviromental factor nor gene mutation factor(ORge,4.767,7.55).⑤Healthy control group compared with the precancerous lesion group synergy index S= 2.53, attributable proportion of interaction AP=0.478.⑥Healthy control group compared with the gastric cancer group, synergy index S=3.432, attributable proportion of interaction AP= 0.615.⑦Smoking, carrying IL-10AG+GG genotype increased the risk of gastric cancer happen OR=2.942 (P=0.016);[CONCLUSION]:Results indicated that there was a relationship between IL-10-1082A/G polymorphism and susceptibility to gastric cancer. And there was statistically interaction between the IL-10-1082 site A/G polymorphism in IL-10 and H. pylori infection in the occurrence and development process of gastric cancer in Hexi area of Gansu Province,China. |
