The Relationship Among IL-10 Gene Polymorphisms, Helicobacter Pylori And Noncardia Gastric Cancer Development In Fujian Province

Objective:1. To study the distribution of interleukin-10 gene polymorphisms in patients with noncardia gastric cancer and patients with chronic superficial gastritis in Fujian Province. 2. To study the relationship among IL-10 gene polymorphisms, Helicobacter pylori (Hp) infection and noncardia gastric cancer development.Method : Patients were divided into two groups,noncardia gastric cancer (NGC) and chronic superficial gastritis (CSG) . Patients with NGC were recruited by age, sex, gastric cancer family history, diagnosis and pathological diagnosis. IL-10 gene polymorphisms analyzed by PCR and direct sequencing. Hp infection was diagnosed by rapid urease test with gastric mucosa ,13C expiration test and serum Hp-IgG test by ELISA.Result: 1. The A/A, A/G and G/G genotype frequencies of IL-10-G1082A were 17.4%,26.4% and 9.4% in NGC patients, 20.9%,10.2% and 15.7% in CSG patients, respectively. There were significant differences of IL-10-1082 gene polymorphism between NGC and CSG.2. The C/C, C/A and A/A genotype frequencies of IL-10-C592A were 44.8%,31.2% and 24% in NGC patients, 41.8%,22.7% and 35.5%, in CSG patients respectively. There were no significant differences of IL-10/-592 gene polymorphism between NGC and CSG .The genotype frequencies of IL-10-819 were same as IL-10/592.3. In NGC, the frequency of genotype GCC/GCC,ATA/ATA and others of IL-10-1082/819/592 were 51.3%,50.8%and 54.7% .In CSG, the frequency of genotype GCC/GCC,ATA/ATA and others of IL-10-1082/819/592 were 48.7%,49.2%and45.3%,respectively. There were no significant differences between NGC and CSG.4.In NGC patients,the frequency of the genotype IL-10-1082G/G,G/A,A/A in Hp(+) was 68.2%,67.7%and 58.5%,respectively; the frequency of IL-10-1082G/G,G/A,A/A in Hp(-) was 31.8%,32.3%and 41.5%.
