Experimental Studies Of JieZe Prescription Gel On Natural Defensive Factors In Mice Vagina

Part One :The Experimental Study of JieZe Gel on Secretory Leukocyte Protease Inhibitor in Mice ViginaObjective:To study the effect of differernt dose of JieZe1 gel.,JieZe2 gel on Secretory leukocyte protease inhibitor(SLPI) in mice vagina.Methods:220 female BALB/ C mice were divided into normal control group and medicine gel group. Medicine gel group, including blank gel,10%Nonxynol-9group,low dose of JieZe1 gel (0.5g/ml),middle dose of JieZe1 gel (1g/ml),high dose of JieZe1 gel (2g/ml),low dose of JieZe2gel(0.5g/ml),middle dose of JieZe2 gel (1g/ml),high dose of JieZe2gel(2g/ml),berberinegroup(0.3mg/ml) and the group consisted of 10%Nonxynol-9 and berberine, was then divided into two sub-groups after administration of 24h and 72h. Each mouse in every group was subcutaneously injected physiological estradiol benzoate (E2) 2μg, and then injected 20μl gel or saline medicine in vaginal respectively after 72h. Each group of 10 mice injected of saline or Gel medicine after 24h, 72h for were killed. The expression of SLPI in vaginal tissue of every group was detected by Western boltting and immunohistochemistry. Results:1. Immunohistochemistrical results: The SLPI in both endochylema and membrane of mice vaginal epithelial tissue express positively,which is showed brown particles under the microscope. The expression of SLPI in vaginal epithelial cells was no significant difference (p> 0.05)between per medicine gel group and the blank control group, ,while the same reslut among the difference of per the medicine gel group (p > 0.05).2.Wstern boltting results: Compared with blank control group, the expression of SLPI in mice vagina tissue of per medicine gel group after 24h and 72h was no significant difference(p > 0.05).Conclusion:When used JieZe1 gel or JieZe2 gel in one time, the expression of SLPI in normal mice vagina has no effect on the vaginal enviroment. Part Two: The Experimental Study of JieZe Gel on Surfactant Protein D in Mice Vagina Objective:To study the effect of the effect of differernt dose of JieZe1gel,JieZe2gel on sufactant protein D(SP-D) in mice vagina.Methods:140 female Kunming mice were divided into normal control group and medicine gel group. Medicine gel group, including blank gel,low dose of JieZe1 gel (0.5g/ml),high dose of JieZe1 gel (2g/ml),low dose of JieZe2gel(0.5g/ml),high dose of JieZe2gel(2g/ml),10%Nonxynol-9group,was then divided into two sub-groups after administration of 24h and 72h. Part Three : The Experimental Study of JieZe Gel on Lactoferrin in Mice VaginaObjective:To study the effect of differernt dose of JieZe1gel,JieZe2gel on Lactoferrin in mice vagina. Methods:100 female Kunming mice were divided into normal control group and medicine gel group. Medicine gel group, including blank gel,10% Nonxynol-9group,berberine group(0.3mg/ml) and the group consisted of 10% Nonxynol-9 and berberine,was then divided into two sub-groups after administration of 24h and 72h. Each mouse in every group was subcutaneously injected physiological estradiol benzoate (E2) 2μg, and then injected 20μl gel or saline medicine in vaginal respectively after 72h. Each group of 10 mice injected of saline or Gel medicine after 24h, 72h for were killed. The expression of LF in vaginal tissue of every group was detected by the method of Western boltting.Results:Wstern bolting results: Compared with blank control group, the expression of LF in mice vagina tissue of per medicine gel group after 24h and 72h was no significant difference(p> 0.05).Conclusion:When used JieZe1 gel or JieZe2 gel gel in a time, the expression of LF in normal mice vagina has no effect on the vaginal enviroment.
