Objective:To observe the effect of capsaicin (CAP) on body fat and serum lipids in alimentary obesity rats and investigate its mechanisms.Method:According to the weight,36 adult male SD rats were divided randomly into six groups:normal diet group (ND), high-fat diet group (HD), capsaicin prevention group (HD+C) and three capsaicin therapy groups:high-fat diet with low capsaicin supplement group (HD+CL), high-fat diet with middle capsaicin supplement group (HD+CM) and high-fat diet with high capsaicin supplement group (HD+CH). The rats in ND group were fed by normal diet and rats in other groups were fed by high fat and carbohydrate diet. Since the fifth week, the HD+C group rats were treated with CAP with a dose of 15 mg·kg-1·d-1 for four weeks, then killed. Since the ninth week, the HD+CL, HD+CM, HD+CH group rats were treated with CAP with the dose of 5 mg·kg-1·d-1,15 mg·kg-·d-1,45 mg·kg1·d-1 respectively for five weeks, then killed. The serum glucose (GLU), total triglycerides (TG), total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C) and the serum visfatin were assayed respectively. And we also observed the pathology changes in liver of the rats.Result:1. After having feed by high fat and carbohydrate diet for eight weeks, the body weight of rats in HD, HD+CL, HD+CM, HD+CH groups, compared with the HD group, were increased by 29.8%,27.6%,29.0% and 28.2% respectively.2. Compared with the HD group, the body weight of HD+C group rats were decreased by 20.7%(P<0.01), and there was no difference between the HD+C group and ND group (P>0.05). The serum TC (P<0.01), LDL-C (p<0.01), visfatin (P0<0.01) were also reduced in HD+C group while the HDL-C (P<0.01) in HD+C group was increased. However, there are no difference in serum GLU and TG between HD+C group and HD group.3. Compared with the HD group, the body weight of rats in HD+CL HD+CM, HD+CH groups were decreased by 7.1%,15.7% and 25.3% respectively, and the weight loss effect of group HD+CH was most obviously, since there was no differences between the HD+CH group and ND group(P>0.05). The wet weight of visceral fat, serum TC LDL-C, visfatin were reduced in HD+CL, HD+CM, HD+CH groups while the HDL-C(p<0.01) in these three groups were increased, and the effect of HD+CH group(45 mg·kg-1·-d-1) is the best. However, there are no difference in serum GLU and TG among all the groups. There were serious fatty liver changes in HD group rats, however, the liver condition in the CAP therapy groups.Conclusion:1. The alimentary obesity rats model can be built by feeding high fat and carbohydrate diet for eight weeks.2. The capsaicin can inhibit the body weight increase, body fat aggregation, serum lipids augment and liver fatty change in overweight and obesity rats, thus the capsaicin has the prevention and treatment function on alimentary obesity, and the effect of HD+CH group(45 mg·kg-1·d-1) is the best.3. The capsaicin can inhibit the serum visfatin aggregation in overweight and obesity rats, and it might prevent and treat the obesity, energy metabolism disorders and fatty liver. |
