Effects Of Zhiyixiao On The Expression Of Visfatin Of NAFLD Rats Mode

Objective:The experiment targets for the adipocytokine of Visfatin,to intervene the NAFLD rat model by the TCM of prescription "ZhiYiXiao".the use of molecular biology,pathology,and other means of observing serum biochemical markers,liver histological changes,especially the changes of the protein and the gene of Visfatin’s expression.To monitor the effect of different concentrations of "ZhiYiXiao" aqueous solution to the expression of Visfatin mRNA and protein,and to explore whether Vifatin as "ZhiYiXiao"target for the treatment of non-alcoholic fatty liver disease and efficacy mechanism.Start with TCM disease Syndromic angle,at the same time using modern medicine,research methods,dialectical debates disease phase combination explore NAFLD the pathogenesis exploration medicine treatment NAFLD efficacy of the mechanism of the new ideas,to provide scientific basis for Chinese medicine treatment of NAFLD,for the development of a provide a theoretical basis for high quality and low price,reliable efficacy of new drugs for the prevention and treatment of fatty liver.Methods:NAFLD models were established in rats by high fat diet.Sixty male adult SD rats in a cleaning grade were randomized into six groups:A,B,C,D,E,F,which were respectively represent the normal control group,sodium chloride negative control group,low dose of treatment groups of ZhiYiXiao,medium dose of treatment groups of ZhiYiXiao and high dose of treatment groups of ZhiYiXiao and YiShanFu capsule positive control group.The Rats in the group B were fed with normal saline each day.And the ones in the groups C,D,E were respectively fed with low,medium and high dose of "ZhiYiXiao",which correspondingly are 3,6 and 12 times of the clinical use for the adult people.And the Rats in the group F were fed with YiShanFu solution,which is 6 times of the dose for the adult people.After 12-week-treatment,the rats were killed,and the blood and liver tissue of them were kept.Series indexes were detected,such as hepatic function,blood fat and liver homogenate.Further,the other pathological change of liver was also investigated through observing the fat variation of the liver cell and the activity of the inflammation.The RT-PCR assay was employed to detect the expressions of Visfatin mRNA,and ELISA assay was employed to detect the expressions of the protein of the Visfatin.Results:It was found that the serological sign of ZhiYiXiao treatment groups and YiShanFu group were significantly decreased and lower than the model group(P<0.01,P<0.05).The Zhiyixiao treatment groups have better effect on improve liver cell inflammation than the model control group(P<0.05);the expression of Visfatin was increased significantly in liver tissue(P<0.01);There was no noticeable difference between the ZhiYiXiao treatment groups and the Yi-Shan-Fu granule group(P>0.05).the liver homogenates TC,TG detect of the ZhiYiXiao treatment groups and YiShanFu groups also compared significantly reduced by the model group(P<0.05);the expression of the protein and mRNA of the Visfatin in ZhiYiXiao treatment groups were significantly lower than the modes group(P<0.01,P<0.05),compared with the YiShanFu group,they had no significant difference(P>0.05).Conclusion:Certain concentration of the ZhiYiXiao can significantly adjustment NAFDL rats lipid metabolism disorders,slow the inflammation of the liver,improve liver function while reducing Visfatin mRNA and protein expression levels in liver tissue caused by the high-fat diet NAFLD therapeutic effects,its efficacy is similar to the YiShanFu but no greater side effects.Of prescription medicine ZhiYiXiao parked on NAFLD within the phlegm of card has a good effect,combined with the clinical situation,addition and subtraction can be widely used in clinical.
