The Effect Of Shaoayo Bawei Decoction On The PCNA & Bax Of Rat With Chronic Atrophic Gastritis

Objective: By observing the general condition, gastric mucosal pathology and the impact of proliferating cell nuclear antigen(PCNA) and B-cell lymphoma/leukemia-associated X protein(Bax) of CAG rats which been treating by Shaoayo Bawei Decoction, to discuss the function mechanism, reveal the pathogenesis of CAG formation, and provide the theory basis for the clinical application.Methods: Use general modeling method to copy the CAG rats model. 110 healthy Wistar rats, male and female was fifty-fifty and feeding 1 week adaptively, in accordance with the random number table, randomly selected 20 as the normal group, the remaining 90 used 0.1% ammonia and 20mmol/L of sodium deoxycholate free alternative to drink, combined with irregular diet method to establish the CAG rats model, a total of 90 days. Confirm the model successfully copied, the remaining 80 rats were randomly divided into model group, Weimeisu group, high-dose group of Shaoyao Bawei Decoction (referred to high-dose group), low-dose group of Shaoyao Bawei Decoction (referred to low-dose group), 20 each group, and male and female was fifty-fifty. The rats were administrated at the same time, the model control group and blank control group were fed with equal volume of normal saline for 30 days. All rats were stopped feeding at least 24 hours after the last administration and then killed by cervical dislocation. Their stomaches were collected to detect PCNA and Bax by enzyme-linked immunosorbent(ELISA), and to carry out HE staining and immunohistochemical staining.Results: (1) Histopathological observation of gastric mucosa: By observing the mucosal morphology, the quantity of lamina propria glands and whether inflammatory cell infiltration or not, we found that the epithelial cells atrophic, degeneration, necrosis or shedding, the number of lamina propria glands significantly reduced and inflammatory cell infiltration in the model group. After Weimeisu,low-dose herb and high-dose herb therapy, gastric atrophy are varying degrees of reversal to the most obvious high-dose group. (2) PCNA contents in gastric tissues: The number of PCNA-positive cells in normal control group was fewer than model control group, mainly located in the proliferative zone, distribution and with weaken-color significantly increased The number of positive cells was significantly increased in model control group, and proliferation of mucosal surfaces with the expansion of full-thickness can be expressed in color deepened. Compared with normal group, PCNA content in gastric tissues of model group was significantly higher, both had significant difference (P<0.01); Compared with model group, Weimeisu, low-dose and high-dose Shaoayo Bawei Decoction could reduce the gastric content of PCNA effectively, with which there was significant difference (P<0.01); In three treating group, PCNA content was no significant difference (P>0.05) among them. (3) PCNA expression characteristics among groups: The PCNA expression in model group was significantly higher than the normal group, both had significant differences (P<0.01);Compared with model group, the number of PCNA was significantly lower in Weimeisu, low-dose and high-dose Shaoayo Bawei Decoction, with whom there was significant difference(P<0.01); Compared low-dose group with high-dose and Weimeisu group, there was significant difference among them (P<0.01); Weimeisu group and high dose group was not significantly different (P>0.05). (4) Bax contents in gastric tissues:Compared with normal group, Bax content in gastric tissue in model group decreased significantly, it had significance (P<0.01); Compared with model group, Weimeisu, low-dose and high-dose Shaoayo Bawei Decoction could Improve the gastric content of Bax effectively, with which there was significant difference (P<0.01); Compared high-dose group with low-dose and Weimeisu group,there was significant difference among them(P<0.01), Weimeisu group and low-dose group was not significantly different(P>0.05). (5) Bax expression characteristics among groups: We could see a large number of positive cells stained dark brown in the gastric mucosa of normal control group, indicating that Bax protein were highly expressed in normal rat gastric mucosa. Model control group can also be found brown positive cells in gastric mucosa, but the expression was significantly reduced compared with the normal control group, and distributed unevenly. Weimeisu group, high dose and low dose group of Shaoyao Bawei Decoction were also can be seen more brown positive cells. The number of Bax expression was between normal control group and model control group, and high dose group was closer to the normal. Bax expression in gastric mucosa in model group was significantly lower than normal group, it had significant difference (P<0.001); High-dose Shaoayo Bawei Decoction could improved the Bax expression of gastric mucosa effectively, compared with model group, there was significant differences (P<0.005).Weimeisu group and low-dose Shaoayo Bawei Decoction group,both compared with model were not significant differences (P>0.005); In three treating group, Bax expression was no significant difference (P>0.005) among them.Conclusions:1 Shaoyao Bawei Decoction can inhibit the excessive proliferation of gastric mucosal cells of CAG rats;2 Shaoyao Bawei Decoction can improve the contents and expressiones of Bax protein of gastric tissues,and the herbs play a regulatory role in apoptosis;3 The experiment provides a scientific basis for clinical application of Shaoyao Bawei Decoction in the treatmentof CAG patients.
