Objective: This experiment through the observation the CAG rat of epidermal growth factor (EGF) and vascular endothelial growth factor (VEGF) content and expression, after Shaoayo Bawei Decoction treated,to discusses the influence of Shaoayo Bawei Decoction and possible mechanism, for the clinical application provide theoretical basis.Methods: Use improved general modeling method to copy the CAG rats model.110 healthy and sexually mature Wistar rats, male and female was fifty-fifty and feeding 1 week adaptively, in accordance with the random number table, randomly selected 20 as the normal group, the remaining 90 used 0.1% ammonia and 20mmol/L of sodium deoxycholate free alternative to drink, combined with irregular diet method to establish the CAG rats model, a total of 90 days. After modeling, six rats randomly selected were sacrificed and underwent pathological examination and found gastric gland atrophy, a part of stomach tissues intestinal metaplasia, indicating that successful model(4 rats were dead accidentally in the process). Confirm the model successfully copied and then all drugs that making model stopped, the remaining 80 rats were randomly divided into model group, Weimeisu group, high-dose group of Shaoyao Bawei Decoction (referred to high-dose group), low-dose group of Shaoyao Bawei Decoction (referred to low-dose group), 20 each group. The rats were administrated at the same time, the model control group and blank control group were fed with equal volume of normal saline for 30 days. All rats were stopped feeding at least 24 hours after the last administration and then killed by cervical dislocation. Their stomaches were collected to detect EGF and VEGF by enzyme-linked immunosorbent (ELISA), and to carry out HE staining and immunohistochemical staining. Results:(1)Gastric general observation:by observing folds,mucous membrane color,and bleeding,we found that the model group plica reduced or disappeared,less mucus,mucous membrane color whitish or gray,and some are accompanied by scattered bleeding points.After weimeisu,low-dose and high-dose Shaoyao Bawei Decoction therapy,gastric mucosal damage most cases are close to normal control group, the most obvious is high-dose group. (2) Gastric mucosa pathology observation: by observing the quantity of gastric mucosa intrinsic glands,exit inflammatory cell infiltration or not, we found that the model group significantly reduced the number of inherent glands,with disorder,and inflammatory cell infiltration. After weimeisu,low-dose and high-dose Shaoyao Bawei Decoction therapy gastric atrophy are varying degrees of reversal, the most obvious is high-dose group. (3) the impact on EGF in blood serum:The model group serum EGF was significantly higher than the normal group,the two have significant differences in comparison(P<0.01); Shaoyao Bawei Decoction low-dose group,high-dose group and weimeisu group can reduce serum EGF content significantly, comparing to model group there is significant difference(P<0.01);The three treating groups serum EGF concen compare with normal group there is significant differences(P<0.01). The high-dose group serum EGF concen compare with weimeisu group there is significant differences(P<0.01); The low-dose group serum EGF concen compare with weimeisu group there is no significant differences(P>0.05); The low-dose group serum EGF concen compare with the high-dose group there is no significant differences(P>0.05). (4) the impact on VEGF in blood serum:The model group serum VEGF was significantly higher than the normal group,the two have significant differences in comparison(P<0.01); Shaoyao Bawei Decoction low-dose group,high-dose group and weimeisu group can reduce serum VEGF content significantly, comparing to model group there is significant difference(P<0.01); The high-dose group serum VEGF concen compare with normal group there is no significant differences(P>0.05),The other treating groups serum VEGF concen compare with normal group there is significant differences(P<0.01).The three treating group serum VEGF content was no significant difference(P>0.05) among them,but the effect of high-dose group the most obvious. (5) The expression of gastric mucosa VEGF:Little VEGF expressed in the normal group,while relatively strong expressed in the model group,the two groups have significantly different(P<0.05). Compared with model group, Shaoyao Bawei Decoction low-dose group,high-dose group and weimeisu group have lower expression of VEGF,the difference was significant(P<0.05). The expression of VEGF in high-dose group compare with normal group there is no significant differences(P>0.05),The other treating groups of VEGF compare with normal group there is significant differences(P<0.05). The expression of VEGF in three treating group was no significant difference(P>0.05) among them,but the effect of high-dose group the most obvious.Conclusion:1 In the CAG pathologic state, EGF high content and VEGF pathogenesis of mediated the high expression, cause gastric mucosa tissue damage.2 Shaoayo Bawei Decoction can obviously decrease serum EGF content of CAG rats, protect the gastric mucosa.3 Shaoayo Bawei Decoction can obviously decrease serum VEGF content of CAG rats, inhibit its expression and inhibit angiogenesis, and thus to produce treatment function to CAG. |
