Effect Of Jianpi Huayu Jiedu Decoction On The Autophagy Protein Of Beclin-1 And LC3B In Rat With Chronic Atrophic Gastritis

Aim To explore the effect of Jianpi Huayu Jiedu decoction on the proliferation of gastric mucosa epithelium and on the autophagy protein beclin-1 and LC3 B in rats with chronic atrophic gastritis.Method In this experiment,CAG rat model was induced by MNNG ethanol compound method.70 male SD rats,aged 4-5 weeks and weighing 100g-130 g,were randomly divided into 10 normal rats and 60 model rats.The rats inthe model group were fed with 120μg/ml-1 MNNG solution and fasted twice a week.On the second day after fasted,30% alcohol solution was infused into the stomach,1ml·100g-1 for each rat.At the end of the 8th week,two rats in the model group were randomly selected.The gastric mucosa at the junction of body and antrum was taken and stained with he,then the early CAG lesions were confirmed under the light microscope.Then,68 rats were randomly divided into model group,folic acid group and high,medium and low dose group of Jianpi Huayu Jiedu Decoction,and they were given 120μg·ml-1 MNNG solution for free drinking.The treatment lasted for 12 weeks.The dosage was as follows: folic acid group 1.8mg·kg-1·D-1;Jianpi Huayu Jiedu Decoction high dose 21.6 g·kg-1·D-1;medium dose 10.8g·kg-1·D-1 and the low dose 5.4g·kg-1·D-1;normal group and model group were given 1ml·100g-1 of normal saline every day.At the end of the experiment,two pieces of gastric mucosal epithelium were taken from the junction of body and antrum of rats.After he staining,the pathological changes were observed under light microscope.Then the expression levels of PCNA,Beclin-1 and LC3 B in gastric mucosal epithelium of rats in each group were detected by immunohistochemistry.The experimental data were processed by spss 22.0 software.Results 1.General situation of rats: rats in the normal group were normal and active.In the model group,the rats were depressed in spirit,passive in response to external stimulation,curled up in groups,decreased in activity,lack of luster in hair color,and serious in depilation.The amount of food and drinking water decreased significantly compared with other groups,and the nutrition was poor,The rats in each treatment group were generally better than those inthe model group,especially in the high dose group of Jianpi Huayu Jiedu Decoction.2.Pathological changes of gastric mucosa: in the normal group,the glands of gastric mucosa were arranged orderly and the cell structure was complete.In the model group,the number of inherent glands in gastric mucosa decreased,the arrangement structure was disordered,the shape was distorted,the polarity of nucleus was weakened,the nucleus was enlarged and deeply stained.After the treatment of Jianpi Huayu Jiedu Decoction,the histopathological abnormalities of gastric mucosa in atrophic gastritis rats were improved in varying degrees.3.PCNA proliferating protein expression: compared with the normal group,the IOD score of PCNA protein expression in the model group increased significantly(P<0.01);compared with the model group,the IOD score of PCNA protein expression in the folic acid group and the Jianpi Huayu Jiedu decoction group decreased significantly(P<0.01);compared with the low dose group,the IOD score of the high dose group and the folic acid group decreased significantly(P<0.01).4.Beclin-1 and LC3 B protein expression: compared with the normal group,the expression of beclin-1 protein in the model group significantly increased the IOD score(P<0.01);compared with the model group,the expression of beclin-1 protein in the high and medium dose groups of Jianpi Huayu Jiedu decoction significantly increased the IOD score(P<0.01 or P<0.05).Compared with the normal group,the IOD score of LC3 B protein expression in the model group increased significantly(P<0.01);compared with the model group,the expression of LC3 B protein in the high and medium dose of Jianpi Huayu Jiedu decoction increased significantly(P<0.01);compared with the folic acid group,the expression of LC3 B protein in the Jianpi Huayu Jiedu decoctionincreased significantly(P<0.01).Conclusion 1.Jianpi Huayu Jiedu Decoction can obviously improve the pathological manifestations of atrophic gastritis rats,such as the reduction of the number of intrinsic glands in gastric mucosa,structural disorder and intestinal metaplasia.2.Jianpi Huayu Jiedu Decoction can reduce the expression of PCNA and inhibit the over proliferation of gastric epithelial cells.3.Jianpi Huayu Jiedu Decoction can up regulate the expression of beclin-1、LC3B,and regulate the level of autophagy.
