Objective:To observe the anti-tumor effects of Compound GuiZhu Capsules (GZ) on the animal lung caner of transplantation, as well as the immune function and expression of PTEN and mdm2 gene. To explore the mechanism of lung cancer in order to better serve the clinical application of GZ on treatment of lung cancer by providing experimental basis for reference.Methods:Established amice model of lung transplantation with method of axillary subcutaneous inoculation.24 hours after the model were completed, weighed and randomly divided the mice into model group, normal animal group, cisplatin(DDP group), cisplatin with high dose of GZ, cisplatin with middle dose of GZ, cisplatin with low dose of GZ, high dose GZ, middle dose of GZ and doses low dose of GZ group.10 in each group. Then the model group and the normal animal group were fed the same volume of distilled water. DDP group were given lmg/kg Intraperitoneal injection (ip),1 time per day for 3 days. Compound GuiZhu capsules were diluted with distilled water, the high-dose group underwent gastric perfusion at 20g/kg (10 times as adult daily dose in clinic), the middle dose at 10g/kg (5 times as adult daily dose in clinic), and the low-dose group were 5g/kg (2.5 times as adult daily dose in clinic),1 time per day for 10 days. On the 11th day, the mice were weighed, exsanguinated and killed, Whose spleens, thymuses and tumor tissues were weighed. And then the spleen index, thymus index, and the inhibition rate were calculated. Double antibody sandwich ELISA method was used to detect the serum TNF-a and IFN-y levels and immunohistochemistry method was used to detect the tumor tissue expression of PTEN, and mdm2 proteins.Results:Compound GuiZhu capsule(GZ)high and middle dose group showed a significant anti-tumor effect (P<0.05), There're no significant difference between GZ high dose group and cisplatin group (P>0.05). GZ high dose group significantly increased the spleen index of the mice of lung cancer(P<0.01), and was able to alleviate the cisplatin inhibition of immune organs. With the GZ dose increased, the serum TNF-a levelof mice of lung cancer was significantly imcreased, especially GZ high dose group was significantly (P<0.001), high dose group of GZ could increase serum IFN-y level of mice of lung cancer. GZ high dose group can increase the expression of PTEN protein (P<0.01) and decreased the expression of mdm2 protein (P<0.01) in lung cancer tissue of mice, what's more, GZ high dose group have the effect of synergy and Efficiency with DDP (P<0.01).Conclusion:Compound GuiZhu capsules can significantly inhibited transplanted lung cancer of animals, and its mechanism may be related to inhibition of PTEN gene inactivation and reduction of mdm2 genes in lung cancer tissue; Compound GuiZhu capsule not only can significantly increase the organ indices of transplanted lung cancer of animals, but also can significantly improve the level of serum cytokines.
