Study On Anti-tumor Action Mechanism Of Compound Sanjin Capsule In A549 Lung Cancer

Objective:To study the pharmacodynamics of compound sanjin capsule on lung cancer A549 by,and to explore its anti-tumor effect and mechanism,and to provide scientific basis for future research on anti-tumor drugs.Methods:To preparation of logarithmic phase A549 lung cancer cells into 1 x107 cells per milliliter of cell suspension,vaccination in 20 male and female left armpit,combination of nude mice to establish animal model of human lung cancer A549 cells,transplantation tumor in nude mice according to body quality after inoculation were randomly divided into four groups,model groups(such as volume of physiological saline),the positive control drug group(2 mg/kg/d cisplatin,IP),the high dose group(4 g/kg/d,ig)and low dose groups(2 g/kg/d,ig),14 day after dosing,compound sanjin capsule in vivo antitumor effect and influence of viscera;ELISA was used to detect the levels of TNF-α,IL-2,IL-12 and IFN-y in the serum of nude mice.The expression of Cyt-c,Caspase-3,Caspase-9,Bax and Bcl-2 in tumor tissues were detected by immunohistochemical staining.The apoptosis of tumor tissue cells was detected by Tunel staining.HE staining was used to observe the histopathology of the tumor tissues.Results:1.The compound sanjin capsule has obvious anti-lung cancer A549 effect in vivo,and the inhibitory rate of the high and low dose group is 49.76%and 33.90%,which has no obvious effect on the relative weight of each organ.2.Compound sanjin capsule can increase serum i1-2,i1-12,TNF-α and IFN-γlevels.3.Immunohistochemical staining results show that compared with model group,Bax positive cells in each dose group of tumor tissue,Cyt C positive cells,caspase-positive cells and caspase 3-9 number of positive cells were significantly increased,the Bcl-2 number of positive cells significantly reduced.4.The results of Tunel staining showed that the Tunel positive and fine running Numbers of compound sanjin capsules were significantly increased compared with the model group.5.Pathological morphological observation was carried out by HE staining,Result display,the tumor cell necrosis could be significantly promoted by the combination of compound sanjin capsule and each dose group.Conclusion:Compound sanjin capsule has obvious anti-lung cancer A549 effect in vivo,and its mechanism may be related to regulating immune imbalance,inducing cell apoptosis and activating mitochondrial apoptosis pathway.
