| OBJECTIVE: To investigate the attenuated effect of Iso and compound paclitaxel on transplanted Lewis lung cancer in C57BL/6 mice and its possible mechanism in vivo.METHODS:(1) Tumor model was set up by transplantating Lewis lung cancer cells into C57BL/6 mice. To investigate the effect of Iso on tumor growth, cell proliferation and apoptosis in transplantation Lewis cell line in C57BL/6 mice. Lewis cells were inoculated into the the C57BL/6 mice to establish Lewis lung cancer model, then thirty C57B1/6 mice with Lewis tumor were randomized three groups: control group, Iso group, Doxorubicin group. After 7 days treatment, samples of tumor were collected. The sections of tumor were observed under light microscope.(2) 84 C57BL/6 mice were randomized into 7 groups, each group contained 12 mice, and then were inoculated subcutaneously with Lewis cells. 24 hours later, Mice in control group were administered intraperitoneally with physiologicsaline, experimental group with paclitaxel, Isoland compound paclitaxel respectively, both in low and high doses continuously (once every day, 5 times altogether ).Mice were killed on sixth day after injection. Tumor weight were measured and tumor-inhibitory rates were calculated; pathology examination: the morphological changes of tumor cells were studies with HE light microscopy; The apoptosis of tumor cells induced by compound paclitaxel and its influence on cell cycle were studied with flow cytometry.RESULT:We observed that the tumor weight was significantly lighter and the tumor size smaller compared to the control group, and the apoptosis of Lewis cells in every group was observed , The cells showed characters of apoptosis and the differentiation was observed by light microscopy. Otherwise, Cell stage was not affluenced obviously.CONCLUSION:The results more confirmed that Iso,Isoland compound paclitaxel could inhibit the growth, proliferation of the transplanted Lewis lung cancer in C57BL/6 mice and induced differentiation of Lewis lung cancer cells, the mechanism may be related with inducing tumor cells apoptosis. The results can provide a guide for laying down the chemotherapy for human tumors, when Iso ,Isol and compound paclitaxel was used as anti-cancer agent. |
