Objective:To investigate the effect of casticin on cell proliferation of human SGC-7901 cell, and it's molecular mechanism.Methods: MTT was used to test the inhibitory effects of casticin on proliferation in SGC-7901 cells. Flow cytometry with FITC labeled annexin V staining was used to determine the percentage of apoptotic cells. the mRNA expression of Bcl-2 and Bax were detected by RT-PCR. And Western Blot was used to analyze the expression of Bcl-2 and Bax.Results: MTT assay showed that casticin significantly inhibited proliferation of SGC-7901 cells in a concentration dependent manner. After treatment with casticin(1.0μg/mL,2.0μg/mL,4.0μg/mL,8.0μg/mL,16.0μg/mL) for 48h, SGC-7901 cells growth inhibit rates were 19.78%,29.79%,47.47%,55.62%,60.32%. Data of flow cytometry of FITC-Annexin-V/PI double staining demonstrated that casticin significantly induced increase of the percentage of apoptotic cells(P<0.05), in a time- and concentration-dependent manner. The RT-PCR and Western blott results displayed that casticin downregulated the expression of Bcl-2 mRNA and protein levels, and upregulated the expression of Bax mRNA and protein levels.Conclusions: Casticin inhibits proliferation of SGC-7901 cells in a concentration dependent manner; Casticin effectively induces apoptosis of SGC-7901 cells, and it's mechanism is associated with downregulation of Bcl-2 and upregulation of Bax in protein and mRNA levels.
