| Objective: By observing the gastric compound containing medicine serum of gastric cancer SGC-7901 cell line proliferation and proliferation of activated receptor gamma peroxidase(PPAR gamma) the effects of gene activity, discuss the stomach compound, possible mechanisms of inhibition of gastric cancer SGC-7901 cell proliferation.Methods: Using blank group,low dose group,middle dose group,high dose group of gastric compound drug plasma to treat stomach gastric cancer SGC-7901 cells,the means of CCK-8 to observe cell proliferation inhibition.After 48 h of using different concertrations of gastric compound drug plasma to treat stomach gastric cancer SGC-7901 cells,use the means of q PCR to detect PPAR gamma expression in gastric cancer SGC-7901 cells.Results:The IC50 of low concentration of stomach drug plasm was 11.1%±2.0%, the middle concentration was 13.5%±3.1%,the high concentration was 12.3%±1.5%. The IC50 of all the serum was not significant difference(p < 0.05),so the research demonstrated that all the concentration of plasma was 10% in effect on SGC-7901 cell line proliferation.The gastric compound containing drug plasma cell proliferation inhibition of SGC-7901, in a time and concentration dependent manner;Stomach compound blank group, low dose group, middle dose group, high dose group of gastric cancer SGC-7901 cell lines after 24 hours OD values were 0.697±0.040,0.654±0.032,0.548±0.048,0.036±0.029;After 48 h OD values were 1.233±0.081,1.131±0.040,0.856±0.055,0.413±0.065;After 72 h OD values were 1.736±0.026,1.362±0.013,0.748±0.024,0.326±0.020, the differences were statistically significant( p < 0.05).PPAR gamma gene expression in gastric cancer SGC-7901 cells rise.After 48 h in concentration dependence. Low dose, middle dose and high dose of stomach compound medicine relative expression of plasma is blank plasma respectively(41.1172±1.9615) times,(4448.5509±4.7522)times,(14893. 2846±1.0381) times,the differences were statistically significant( p < 0.05).Conclusion: 1,The drug plasma of gastric compound with the increase of drug concentration as a function of time extension on gastric cancer cell line SGC-7901 proliferation inhibition increased gradually, in a time and concentration dependent manner.It suggests that gastric compound has inhibitory effect on the proliferation of gastric cancer. 2,After the intervention of gastric compound drug plasma, SGC-7901 cells PPAR gamma expression level was dose dependently increased, suggesting that gastric compound containing plasma may be inhibited PPAR gamma gene expression through the way to suppress the proliferation of SGC-7901 cells. |
