[Objective] To investigate the expressions of Cripto-1,Nodal and Smad2 in normal cervical epithelium,cervical intraepithelial neoplasia,invasive carcinoma of cervix,and the relationship between progression of disease and it,and to investigate the significance.[Methods]①Western blot was used to examine the expression levels of Cripto-1,Nodal in the 26 cases of cervical carcinoma,the 52 cases of cervical intraepithelial neoplasia and the 20 cases of normal cervical epithelium.②RT-PCR was used to examine the expression levels of Cripto-1,Nodal and Smad2 in the 26 cases of cervical carcinoma,the 52 cases of cervical intraepithelial neoplasia and the 20 cases of normal cervical epithelium.③Immunohistochemistry method was used to examine the expression levels of Cripto-1,Nodal in the 26 cases of cervical carcinoma,the 52 cases of cervical intraepithelial neoplasia and the 20 cases of normal cervical epithelium.The association between expression of Cripto-1,Nodal with clinicopathologic parameters was assessed.④Using immunohistochemistry method,we examined Cripto-1,Nodal expression levels of Caski cells before and after treatment.[Results]①Western-blot showed that the average expression level of Cripto-1,Nodal in ICC was obviously higher than in CIN and NCE respectively(Cripto-1:0.92±0.68 vs. 0.31±0.23 & 0.09±0.06,P<0.05;Nodal:0.79±0.40 vs. 0.30±0.23 & 0.10±0.08,P<0.05) ,and it in CIN was higher than in NCE respectively(P<0.05).②RT-PCR revealed the average expression level of Cripto-1 mRNA,Nodal mRNA,Smad2 mRNA in ICC was higher than in CIN and NCE respectively(Cripto-1:0.62±0.36 vs. 0.24± 0.14 & 0.10±0.09,P<0.05;Nodal:0.86±0.39 vs. 0.38±0.30 & 0.14±0.12,P<0.05;Smad2:0.85±0.39 vs. 0.35±0.25 & 0.19±0.17,P<0.05) ,and it in CIN was higher than in NCE respectively(P<0.05).③The immunohistochemistry method displayed the positive expression rate of Cripto-1,Nodal was 57.7%(15/26),84.6%(22/26) respectively in ICC,but significantly lower in CIN(26.9%,30.8%)and NCE(5.0%,10.0%)(P<0.05).Cripto-1 protein expression in ICC was correlated to lymph node metastasis(P<0.05),but it was no correlation to patients'age,histological type,pathologic grade,clinical stage(P>0.05);Nodal protein expression in ICC was correlated to pathologic grade,lymph node metastasis(P<0.05),but it was no correlation to patients'age,histological type,clinical stage(P>0.05).④Cripto-1,Nodal expression brought down obviously in Caski cells after treatment.[Conclusion]1. The expression level of Cripto-1,Nodal obviously increased in cervical carcinoma,there was a significant difference between normal cervical epithelium and it,and there was significant relationship between Cripto-1,Nodal expression and pathologic grade,lymph node metastasis,it displayed Cripto-1 and Nodal maybe play an important role in the carcinogenesis and development of cervical carcinoma.2. Cripto-1 and Nodal could be used as a tumor marker for early diagnosis of cervical carcinoma and precancemns lesions.3. Cripto-1,Nodal expression brought down obviously in Caski cells after treatment,suggested Cripto-1 and Nodal could become a target for evaluating cancer therapeutic effect. |
