Background and Objective:As science and technology development and people's increasing awareness of cancer through the detection of various tumor-related tumor markers for early diagnosis and prognosis of cancer surveillance became a hot topic in cancer research today. PTEN gene is the first of a dual-specificity phosphatase activity of tumor suppressor gene,with control cell growth and apoptosis,and can affect the tumor cell invasion and metastasis process. Cyclooxygenase (COX-2) is prostaglandin (PG) biosynthesis in the process of an important enzyme, can be posted arachidonic acid metabolism for various PG product, thus maintaining the body in the pathophysiological process.Both with tumor invasion, metastasis and angiogenesis are closely related, multi-step evolution of the tumor plays an important role, but which angiogenesis is a tumor cell survival, nutrition and access to the most basic conditions of distant metastasis.There is few report of the research of OSCC.Hence, this article adopts streptavidin-biotin peroxidase(S-P) method to examine normal oral mucosas (NOM), oral squamous cell carcinoma (OSCC). To investigate expression of phosphatase and tensin homologue deleted on chromosome ten and cyclooxygenase-2 and their relationship with angiogenesis in oral squamous ce1l carcinoma.Methods:1. Choose 10 examples of NOM and 50 examples of OSCC that is of entire clinical materials, and collect the materials of age,sex,lymph grading ,lymph node diversion.2. Adopt SP method to investigate make clinical stage of the 10 examples of NOM , and 50 examples of OSCC ,to find the expression of PTEN,COX-2. Microvessel Density (MVD) in OSCC was as-sessed with microscope.3. Statistically analyze the difference of NOM and OSCC of PTEN and COX-2 ,analyze PTEN and COX-2's expressing and the relation among clinical stage of OSCC,pathological grading and lymph node diversion ,and the relation of PTEN,COX-2 and MVD.Result:The positive rates of PTEN and COX-2 expression were 100% and 0.0% in nomal mucosa, 56.0% and 58.0% in OSCC tissue. MVD was 45.63±10.52. The PTEN expression was significantly associated with differentiation and clinical stages (P<0.05). The COX-2 expression was significantly associated with differentiation ,clinical stages and lymph node metastasis status (P<0.05).In the positive expression cases of PTEN MVD was 41.17±10.46,in the negative expression cases of PTEN MVD was 47.46±12.31,there was significant difference in two groups(P<0.05).In the positive expression cases of COX-2 MVD was 45.32±12.87,in the negative expression cases of COX-2 MVD was 37.18±9.68,there was significant difference in two groups(P<0.05). The expression of PTEN was negative correlated with COX-2 (rs=-0.509,P<0.05).Conclusion:PTEN inactivation and COX-2 overexpression with angiogenesis play an important role in tumor progression in OSCC. Combination measurement of PTEN,COX-2 and MVD will help to judge the diagnosis and prognosis of OSCC.
