Objective:To study of the oral Candidal colonization status with different base-materials denture. To compare the adhesion of Candida albicans to two different dental restorative materials in different incubation condition (static and shaking incubation), and study the effect of vibration frequency on the adhesion.Methods:Non-stimulated saliva were collected by standard procedure as samples by 147 cases who wear dentures. Salivary samples were inoculated onto CHROMagar CandidaTM to identified Candida strains. The total number of colonies was considered as the detectable intensity of oral Candida. Statistical analysis was used to compare with the differences of detection rate and detection intensity.The columniform specimen made of dental Co-Cr alloy and dental Titanium alloy were polished step by step. Then the columniform specimen were separate randomly put into liquid Sabouraud culture medium in which there are Candida albicans and incubate in the condition of different vibrating speed for 24h. Afterwards the adhesive microbial amount were determined by the clone forming unit counting method with CHROMagar CandidaTM, and adhesion morphology was analyzed with Fluorescence microscope.Results:Candida was isolated from 147 cases who wear different base materials denture, such as Candida albicans. Candida tropical, Candida krusei. The frequency showed significantly difference between Candida albicans and non-Candida albicans(P<0.05). The frequency showed no difference between two strains of non-Candida albicans(P>0.05).The frequency of Candida showed no difference between group A. B. and C (P>0.05). The Candida albicans CFU count was significantly higher in group A than in group B and C (P<0.05); group B was significantly higher than group C (P<0.05). The non-Candida albicans CFU count showed no difference between three groups (P>0.05).In two different incubation conditions, the adhesion of Candida albicans were significantly difference between the same columniform specimen (P<0.05). In the shaking incubation conditions, there are more adhesive microbe on both materials by stationary incubation method that by shaking incubation method:and the differences are statistically significant (P<0.05).Conclusion:Candida albicans is main strains in oral Candidal. the other two Candida strains were significantly lower than the proportion of candida albicans exists in activity in the oral denture wear.Three strains of Candida albicans, Candida tropicalis, Candida krusei can be detected in people who wear different base material denture. CFU count of oral candidiasis are closely related to the use of denture base materials, titanium and titanium base alloy denture are the best choice to prevent denture stomatitis.The adhesive microbe of Candida albicans on Co-Cr alloy was significantly higher than Titanium alloy in same incubation condition method. The adhesive microbe of Candida albicans by shaking incubation method was significantly lower than static incubation method.
