objective:(1)Use standardized psychological tests, to assess the personality characteristics of anxiety disorders objectively.(2)Use the advanced surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI - TOF -MS ) to search the specificity of protein fingerprint and the proteomics characteristics for anxiety disorders, and then analyzes its structure, properties and quantity changes, to reveal the etiology and pathogenesis of anxiety disorders from protein molecule level.(3)Use the advanced mathematical statistical analysis related technologies, to explore the interaction law and its relevance on mechanism between the protein groups and personality characteristics; constructing the pathogenesis of anxiety disorders; promote the research of the pathogenesis and the etiology of anxiety disorders from comprehensive empirical model to a more in-depth comprehensive thinking phenomenology of biological - psychological - social culture etiology comprehensive empirical research, thus pushing the treatment from simple treatment for symptomatic treatment etiology and pathogenesis to the fundamental treatment direction. Methods : (1)according to the America diagnostic and statistical manual of mental disorders the fourth edition of the diagnostic standard (DSM -Ⅳ) , choose 31 anxiety disorders patients (including the extensiveness anxiety disorders, panic disorders, panic, compulsive disorder and post-traumatic stress disorder, etc) from the psychological outpatient service of the Luzhou Medical College as the experimental group, and also establish the control: random normal crowd 54 people;(2) Use the Minnesota Multiphasic Personality Inventory (MMPI), to measure and analysis the personality characteristics of the experimental group and the control group respectively (.3)Using the PBSII/C protein fingerprint apparatus and Protein chip CM10 (weak cation exchange surface) produced by Ciphergen company cluster the protein microarray of the experimental group and the control group, using the analysis software Biomaker Wizard3.2 , to screening the differences protein .Then receiver operating characteristic (ROC) curves were used to appraise the distinct proteins. Finally the Fisher's discriminant model was established to reveal the etiology and pathogenesis of anxiety disorders in protein molecular level.Results: (1) MMPI:①the score (T points) in the following factors exist significant differences between two groups: F,K,D,Hy,Pd,Pa,Pt,Sc,Ma,Si(p<0.05 or p<0.01);②the score (T points)between male patients and female patients in the following factors exist significant differences: Hs,D,Hy,Pd,Mf,Pt(p<0.05 or p<0.01);⑤the Two point Codes of the experimental group:the majority of patients present the personality type with 78/87,27/72,28/82(P>75%)。⑵Relatively 4 proteins were lowly and 92 proteins highly expressed between AD patients and healthy controls. All of the 96 distinct proteins were analyzed by ROC. 8 proteins (AUC>0.75) were selected to establish the Fisher's discriminant model and 3 proteins (m/z: 2612,4484,5640) were accepted by the model. The accuracy of AD from healthy controls was 86.6%, the sensitivity and the specificity were 85.7% and 87.0%, respectively. (3)After logistic regression analysis ,there are four factors :Pt(OR=1.772,95%CI:1.145~2.741),Sc(OR=0.767,95%CI:0.604~0.972),M4484(OR=1.316,95%CI:1.056~1.639),M5641(OR=1.820,95%CI:1.175~2.820) were accepted by the model.Conclusion:(1) There is certain personality of patients with anxiety disorder: neuroticism, psychoticism and social introvertion, etc. The manifests are sensitive suspicious,depression, hypochondria, self-centered, lack of self-confidence, social withdrawal, etc. (2) The main personality types of anxiety disorders are 78/87, 27/72, 28/82 etc (3) The differentially expressed serum molecular proteins have a high diagnostic specificity for AD from healthy individuals. (4)Using the SELDI - TOF - MS technology to test anxiety disorders serum features at the molecular level, provides a new method for the study of pathogenesis and clinical diagnosis of anxiety disorders, as well provides a objective experimental index of anxiety disorders, and deserves a further study. (4)Using the logistic regression analysis, the prediction result of the model established for anxiety disorder is 89.7%. (6)Anxiety disorders is the result of joint action of psychological factors with biological factors, and biological factors is more important in its occurrence and development. |
