Objective:To observation the effect of rhubarb given at different time during the perioperative period on intestinal mucosal barrier in rats after partial hepatectomy, and to investigate the mechanism of effect. Methods:One hundred and fifty healthy male SD rats were randomly divided into four groups:control group, model group, preoperative administration group, intraoperative administration group and postoperative administrati-on group, Rats were sacrificed at different time(1 d,3 d,7 d,)after partial hepatectomy, superoxide dismutase SOD levels in serum and ileum tissue were determined, Maleic Dialdehyde MDA levels in serum and ileum tissue were determined and the density of endotoxin in serum and ileum tissue were determined the change of ileum tissue pathology was observed by optics microscope. Chiu's count was used to assess the changes in intestinal mucosal pathological morphology. Results:1)In the 1d,3d after partial hepatectomy, compared with control group, the activity of SOD in serum and in ileum tissue was decreased in model group, preoperative administration group, intraoperative administration group and postoperative administration group, the differences were significant(P<0.05), the content of MDA in serum and in ileum tissue was increased, the differences were significant(P<0.05), the density of endotoxin in plasma and in ileum tissue was increased, the differences were significant(P<0.05), the Chiu's count was increased, the differences were significant(P<.05). Compared with model group, the activity of SOD in serum and in ileum tissue was increased in preoperative administration group, intraoperative administration group and postoperative administration group, the differences were significant(P<0.05), the content of MDA in serum and in ileum tissue was decreased, the differences were significant(P<0.05), the density of endotoxin in plasma and in ileum tissue was decreased, the differences were significant(P<0.05), the Chiu's count was decreased, the differences were signifi- cant(P<0.05);compared with preoperative administration group, the activity of SOD in serum and in ileum tissue was increased in intraoperative administration group and postoperative administration group, the differences were significant(P<0.05), the content of MDA in serum and in ileum tissue was decreased, the differences were significant(P<0.05), the density of endotoxin in plasma and in ileum tissue was decreased, the differences were significant(P<0.05), the Chiu's count was decreased, the differences were significant(P<0.05); 2) In the 7 d after partial hepatectomy compared with control group, the activity of SOD in serum and in ileum tissue was decreased in model group, preoperative administration group, the content of MDA in serum and in ileum tissue was increased, the differences were significant(P<0.05), the density of endotoxin in plasma and in ileum tissue was increased, the differences were significant(P<0.05); 3) The activity of SOD, the content of MDA and the density of endotoxin and the Chiu's count in intraoperative administration group and postoperative administration group, the differences were no significant(P>0.05). Conclusion:Using rhubarb in different time in the perioperative period can protect the intestinal mucosal barrier of partial hepatectomy rats by increasing the activity of SOD in serum and in ileum tissue, decreasing the content of MDA in serum and in ileum tissue, decreasing the density of endotoxin in plasma and in ileum tissue and promoting the intestinal mucosal regeneration, However, it showed obvious effect when the treatment was taken at intraoperative administration group and postoperative administration group. |
