Objective: In order to guide a better clinical choice of surgical suture and provide a theoretical basis for clinical treatment, the study that based on an animal model of ageing attempts to research the effects of Teflon suture and Catgut on the skin sutures morphology and the skin aging factors in the operation area. Methods: To establish twenty ageing rabbit model by intra-peritoneum injection of D–galactic. Twenty paired rabbits were randomly divided into two groups. One is Teflon suture group, the other is Catgut group (Group C&D).Each of them has control group (Group K) for comparison. Each rabbit's back was divided into two sides according to spine, and each side was further subdivided into four regularly spaced parts. Teflon suture and Catgut were implanted under the operation area of rabbit skin respectively. Cut skin samples into size of 5mm×5mm×3mm from surgical areas and original areas by the 3rd, 7th, 15th, 30th, 45th, 60th and 90th day of implant. Separate the sample into two copies. One was done HE to observe the histological changes on operation area of skin. The other was made into tissue homogenate to measure superoxide dismutase (SOD) activity, hydroxylproline(HYP) content, total antioxidant capacity (T-AOC) and hydrogen peroxide (H2O2)content.Results:Determination of SOD activity: SOD values of Teflon suture group and Catgut group are higher than value of control group at different time points, and value of Catgut group is higher than Teflon suture group at different time points. Differences between any two groups is statistically significant (P<0.05).Determination of HYP content: HYP values of Teflon suture group and Catgut group are higher than value of control group at different time points, and value of Catgut group is higher than Teflon suture group at different time points. Differences between any two groups is statistically significant (P<0.05).Determination of T-AOC: T-AOC values of Teflon suture group and Catgut group are higher than value of control group at different time points, and value of Catgut group is higher than Teflon suture group at different time points. Differences between any two groups is statistically significant (P<0.05).Determination of H2O2 content: H2O2 values of Teflon suture group and Catgut group are lower than value of control group at different time points, and value of Catgut group is lower than Teflon suture group at different time points. Differences between any two groups is statistically significant (P<0.05). Conclusion: Superoxide dismutase activity, hydroxyproline content and total antioxidant capacity of Teflon suture group and Catgut group are higher than control group, and hydrogen peroxide content is lower than control group. This research result maybe show that implanted suture material in the skin of rabbits is possible to stimulate the skin for putting off skin-ageing. At the same time, superoxide dismutase activity, hydroxyproline content and total antioxidant capacity of Catgut group are higher than Teflon suture group, and hydrogen peroxide content is lower. This result maybe show that Catgut is more useful than Teflon suture for putting off skin-ageing. |
