Objective To investigate the association between Apolipoprotein E (ApoE) genotype and children cerebral palsy (CP) in Guangxi and identify the disease-related genotypes or high-risk allele of CP, which will provide gene information to the pathogenesis study of CP .Methods (1)A case-control study was performed which enrolled 83 children with CP who were diagnosed in the First Affiliated Hospital of Guangxi Medical University and l20 children without nervous system diseases. (2) Genomic DNA was extracted from peripheral venous blood leukocytes using phenol chloroform methodology. The ApoE polymorphism was determined by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) and then was validated by DNA sequencing. (3) The ApoE genotype and allele frequency statistics were performed by SPSS 17.0 statistical software using chi-square test.Results Total five genotypes were identified exceptε2/2. In CP group genotype ofε2/3 could be found in 4 cases andε2/4 genotype in 1 case,ε3/3 in 55 cases,ε3/4 in 21 cases,ε4/4 in 2 cases; In control group, genotype ofε2/3 could be found in 14 cases,ε3/3 in 90 cases,ε3/4 in 15 cases,ε4/4 in 1 case. The most common genotype in two groups wasε3/3,others areε3/4 andε2/3. ApoE genotype frequency of CP group:ε2/3 4.82%,ε2/4 1.20%,ε3/3 66.27%,ε3/4 25.30%,ε4/4 2.41%,each allele frequency wereε2 3.01%,ε3 81.33%,ε4 15.66%; In control group, each gene frequency were:ε2/3 11.67%,ε3/3 75.00%,ε3/4 12.50%,ε4/4 0.83%, frequency of allele were:ε2 5.84%,ε3 87.08%,ε4 7.08%,frequency ofε3/4 genotype in CP group was significantly higher in control group(χ2 = 5.511, P = 0.025); frequency of ApoE alleleε4 in CP group was significantly higher in control group (χ2 = 7.627, P = 0.008).Incidence of relative risk(OR value) of ApoE alleleε4 and CP was OR=2.436,95%CI(1.276-4.652).Conclusion ApoE genotypeε3/4,ε4 allele may relate to and CP andε4 allele may be a genetic susceptibility factor. |
