Angelica dahurica Benth.et Hook.f.var. formosana Shan et Yuan belongs to the Umbelliferae family, Angelica genus.Radix Angelicae dahuricae, the roots of Angelica dahurica (Fisch.ex Hoffm.) Benth. et Hook. F. or Angelica dahurica dahuricae (Fisch.ex Hoffm.) Benth.et Hook. F. var. formosann (Boiss.) Shan et Yuan, have been used in Chinese traditional medicine as an analgesic, antibiotic and anti-inflammatory agent. Modern studies have found that Angelica dahurica can anti-inflammatory and analgesic effectively.Objective:Find a method of the quality control and evaluation of Angelica dahurica, which is mainly based on the correlation of the chemical fingerprints and biological effects. And this method can be used for the quality evaluation of medicinal materials from different producing areas.Methods:To separate and purify nine compounds of Angelica dahurica by fractional crystallization and C18 silica gel column chromatography. To determine the contents of six major coumarins and establish the chemical fingerprint of Angelica dahurica from different producing areas by RRLC-UV. The method of grey related analysis was used in the relational analysis of inhibit PLA2 activity and each peak, based on RRLC-UV fingerprints.Results:Nine compounds of Angelica dahurica (β-sitosterol, bergapten, imperatorin, isoimperatorin, phellopterin, cnidilin, oxypeucedanin, oxypeucedanin hydrate, osthole, phellopterin) have been isolated and purified as reference substance for chemical analysis and screening of the active ingredients. The contents of six major coumarins and the chromatographic fingerprints of the different samples have been analyzed and investigated by RRLC-UV. Their similarity of correlation points out the difference of the chemical characteristics of Angelica dahurica, which shows that those chemical characteristics could be applied in the quality control and evaluation of Angelica dahurica. Different processing methods of Angelica dahurica solution can inhibit PLA2, IC50 of dryness under sun and dryness by sulfur are 3.36 mg/ml and 4.51 mg/ml, It is showed that bioactivity of inhibit PLA2 of dryness under sun is better than that of dryness by sulfur. The comprehensive analysis shows that common components relate with the bioactivity of inhibit PLA2 at different extent. The order of grey correlation degree is peak 13 (bergapten)> peak 7 (oxypeucedanin hydrate)> peak 19>peak3>peak 23>peak 10>peak 11>peak 9>peak 18> peak 20 (Cnidilin)> peak S22 (isoimperatorin)>peak 14>peak 1>peak 21 (osthole)>peak 8>peak 17 (imperatorin)>peak 3>peak 2>peak 12>peak 4> peak 5>peak 6>peak 15>peak 16 (oxypeucedanin).Conclusion:Though collection areas and environment of Angelica dahurica are different, the chromatographic fingerprints are similar. There are obvious discrepancies in the relative contents of components. Based on the chemical composition contents together with bioactivity of inhibit PLA2, the result of comprehensive evaluation indicates that Angelica dahurica from Suining and Dazhou in Sichuan province are better than others.
