[Objective] Observe the effect of Yuxianling Granula on the learning and memory ability of kainic acid (KA)-induced epileptic rats and hippocampal neuronal apoptosis,and to explore its mechanism.[Method] Adopting the stereoscopic directional technology to inject the KA,we established rat model of temporal lobe epilepsy.The learning performance and memory of the rat water maze tests was carried out to evaluate the effect of KA on the learning and memory ability of epileptic rats;Using DNA original place end labelling method (TUNEL) to check the cell apoptosis in the hippocampus, using immunohistochemistry to detect the expression of apoptosis-related proteins in hippocampus called Bax and Bcl-2,then we explored the possible way in which Yuxianling Granula exerted the effect on the impaired learning and remembering capacity of epileptic rats.[Results]1 The results of a water maze testCompared with the Blank group,the learning and memory performance of model group was significantly different(P<0.001),it prompted that epilepsy could lead to a decline in learning and memory in rats.Compared with the model group,the learning and memory performance of Yuxianling Granula group was significant different;Compared with the Blank group two indicators was no significant difference (P>0.05),it prompted that Yuxianling Granula could improve the learning and memory damage of epileptic rats significantly.2 The test result of hippocampal neuronal apoptosisAmong each group,test results of the number of apoptotic cells in hippocampus slices was statistically significant difference (P<0.05).Between the Blank group and model group,the difference was statistically significant (P<0.05),Compared with the Blank group,Yuxianling Granula group,lamotrigine group, sham operation group were no significantly different;Compared with model group,Yuxianling Granula group and lamotrigine group were significantly different(P<0.05);The difference between Yuxianling Granula group and the lamotrigine group was not statistically significant (P>0.05).It prompted that epilepsy can lead to hippocampal neuronal apoptosis, Yuxianling Granula and lamotrigine can significantly reduce the number of hippocampal neuronal apoptosis.3 The test results of Bax expressionAmong each group,the test results of the number of Bax positive cells in hippocampus slice was different significantly(P<0.05). Between the Blank group and model group, the difference was statistically significant (P<0.05),Compared with the Blank group,Yuxianling Granula group,lamotrigine group,sham operation group were no significantly different; Compared with model group,Yuxianling Granula group and lamotrigine group were significantly different(P<0.05);The difference between Yuxianling Granula group and the lamotrigine group was not statistically significant (P>0.05).It prompted that epilepsy can increased Bax protein expression in rat hippocampus,Yuxianling Granula and lamotrigine can significantly reduce Bax protein expression in the hippocampus.4 The test results of the expression of hippocampal neurons Bcl-2Among each group,the test results of the number of Bcl-2 positive cells in hippocampus slice was different significantly(P<0.05).Compared with the Blank group, model group,Yuxianling Granula group,lamotrigine group,sham operation group were no significantly different(P>0.05);Compared with model group,Yuxianling Granula group and lamotrigine group were significantly different(P<0.05);The difference between Yuxianling Granula group and the lamotrigine group was not statistically significant (P>0.05).It prompted that epilepsy can not obviously increased the expression of Bcl-2 protein in hippocampus,Yuxianling Granula and lamotrigine can improve the expression of Bcl-2 protein in hippocampus of epileptic rat.[Conclusion]1 The learning and memory capacity of epileptic rats had been impaired by the epilepsy.2 The damage to learning and memory ability caused by Epilepsy may be related to hippocampal neuronal apoptosis.3 Yuxianling Granula can improve learning and memory in rats with epilepsy.4 The mechanism of Yuxianling Granula improving learning and memory ability in rats with epilepsy may be to reduce hippocampal neuronal apoptosis after seizures,this mechanism may be related to enhanced expression of Bcl-2 protein and reduced expression of Bax protein in epileptic rat hippocampal neurons.
