The Effect Of Yuxianling Decoction And Its Flavoring On TLR4-mediated MyD88-dependent Pathway In Rats With Pentylenetetrazol-induced Epilepsy

Objective To investigate the influence of Yuxianling Decoction on the expressions of TLR-4,MyD88 and IL-1? in hippocampus of epileptic rats,and to explore the mechanism of inflammatory response in epilepsy.Methods 130 male healthy SD rats were randomly divided into control group(n=10), sham-operation group(n=10),with normal saline injected in the belly. The other rats were intraperitoneally injected with pentylenetetrazole (PTZ) (n=110). The standarded epileptic rats were randomly divided into model group, valproate group,Yuxianling Decoction(YXL), Yuxianling with honeysuckle Decoction(YXLH), each group containing 10 rats.The expression of TLR4 mRNA protein in rat hippocampus were detected by RT-PCR, the expression of MyD88 and IL-1? protein were tested by immunohistochemical method.Results1.The expression of TLR4 mRNA, MyD88 and IL-1? was no significant differences between control group and sham-operation group (P>0.05);2. Compared with the normal group and sham-operation group,the expression of TLR4 mRNA, MyD88 and IL-1? increased more significantly in model group. (P<0.01);3.Compared with the model group,the expression of TLR4 mRNA, MyD88 and IL-1? was significantly reduced in YXL, YXLH and valproate group.4.The expression of TLR4 mRNA, MyD88, IL-1? in YXL and YXLH was much weaker than that in valproate group (P<0.01);The difference compared with YXL and YXLH was not obvious (P>0.05).Conclusion The increased expression of TLR4 mRNA, MyD88 and IL-1? in rats hippocampus may play an promotion role in epileptic;Yuxianling Decoction can down regulate the expression of TLR4 mRNA MyD88 and IL-1? protein and lessen the pathologic changes of hippocampus. It's no significant differences between YXL and YXLH (P>0.05),we think they may certain reduce inflammatory response in brain after seizures.
