Objective: To study targeting distribution of flurbiprofen axetil in uterine leiomyoma and it's effect on PGE2 and IL-6; To investigate the mechanism of flurbiprofen axetil.Method: 40 cases of ASA 1~2 female patients undergoing elective surgery of uterine leiomyoma were randomly divided into four groups:control group(K0 group),different flurbiprofen axetil injected groups(K1,K2,K3 groups),n=10.K0 group were injected physiological saline, the other 3 groups were injected flurbiprofen axetil 0.5 mg·kg-1,1 mg·kg-1,1.5 mg·kg-1(K1,K2,K3 group) respectively 15 minnutes before skin incision.when the tumor tissue incised, record the time and draw the blood ,incise the tumor tissue and normal tissue 3cm distance.Flurbiprofen of plasma and tissue homogenate were detected by HPLC; PGE2 and IL-6 were detected by ELISA. Analyze these indexes by SPSS13.0.Result:(1)The control group(K0 group) has no flurbiprofen.The plasma flurbiprofen in K1,K2,K3 groups increased along with the dose, the differences were significant(P<0.05).(2)The tissue homogenate of K0 group has no flurbiprofen. The flurbiprofen of tumor tissue homogenate in K1,K2,K3 groups increased along with the dose, the differences were significant(P<0.05). The flurbiprofen of tumor tissue homogenate in K1,K2,K3 groups increased compared to that of normal tissue homogenate, the differences were significant(P<0.01). The flurbiprofen of normal tissue homogenate in K1,K2,K3 groups were not significant(P>0.05).(3)The plasma PGE2 in K0 group was higest. The plasma PGE2 in K1, K2,K3 groups decreased along with the dose, the differences between K0 group were significant(P<0.01). The differences of K2,K3 groups between K1 group were significant(P<0.01),but the difference between K2 and K3 was not significant(P>0.05).(4) The PGE2 of tumor tissue homogenate in K0 group was higest. The PGE2 of tumor tissue homogenate in K1,K2,K3 groups decreased along with the dose,but the difference between K1 and K0 was not significant(P>0.05). The differences of K2,K3 groups between K0 group were significant(P<0.01); the difference between K2 and K1, K3 and K2 was not significant(P>0.05). The PGE2 of tumor tissue homogenate in K0,K1,K2,K3 groups increased compared to that of normal tissue homogenate, the differences were significant(P<0.01). The PGE2 of normal tissue homogenate in K0,K1,K2,K3 groups were not significant(P>0.05).(5) The plasma IL-6 in K0 group was higest. The plasma IL-6 in K1,K2,K3 groups decreased along with the dose, the differences between K0 group were significant(P<0.01). The differences of K2,K3 groups between K1 group were significant(P<0.01),but the difference between K2 and K3 was not significant(P>0.05).Conclusion:(1)The distribution of flurbiprofen axetil in tumor tissue was targeted.(2) Flurbiprofen axetil administration could decrease the expression of pain excitatory transmitter PGE2 in plasma. Flurbiprofen axetil administration of high-dosage(≥1 mg·kg-1)could decrease the expression of PGE2 in tumor tissue. (3)The expression of plasma IL-6 was suppressed by flurbiprofen axetil administration.
