Objectve: Compare the analgesic effect of flurbiprofen axetil on incisional pain in rats bygiving different doses of flurbiprofen axetil at two time points,discuss the preventiveanalgesic effect of flurbiprofen axetil .(1)To observe the effect of preemptive analgesia by injection different doses of flurbiprofenfrom the tail vein on the rat model of incisional pain;(2)Compare the analgesic effect of the two different point (before and after surgery);(3)To provide the basis for preemptive analgesia for the clinical use of flurbiprofen axetil.Methods: 64 male SD rats were randomly assigned to 8 groups (n = 8). The experimentwas divided into two main parts (F and C). F part and C part each contained four groups,saline group (F1, C1 group), flurbiprofen axetil 1,5,9 mg / kg groups respectively(F2, F3, F4group, C2, C3, C4 group ). F part: 20min before the operation were given saline or differentdoses of flurbiprofen axetil . C part: 20min after the operation were given saline or differentdoses of flurbiprofen axetil . Use cumulative pain score test to evaluate the pain behavior, and2 hours later,kill the 9rats,remove the spinal cord and examine the fos expression by usingimmunohistochemistry method .Results: (1)F part: Compared with F1 group , cumulative pain scores of F2, F3 and F4groups were lower (P<0.05), c-fos positive cells decreased(P<0.05); (2)C part : Comparedwith the C1 group, cumulative pain scores of C2, C3 and C4 groups were lower (P <0.05),c-fos positive cells decreased(P<0.05); (3)the comparison of the cumulative pain score of Fpart and C part : there is no significant difference (P> 0.05) between F2 group and C2 group,F3 group and the C3 group , while the F4 group and the C4 group difference was significant(P <0.05), the cumulative pain score of F4 group was lower;(4)the comparison ofimmunohistochemistry of F part and C part : c-fos positive cells were no significantdifference in the number (P> 0.05) between F2 group and C2 group,F3 group and the C4group while the F3 group and C3group, F4 group and C4 group,difference was significant (P<0.05).the number of c-fos positive cells of F3 group is lower,the number of c-fos positivecells of F4 group is lower.Conclusion:(1) Flurbiprofen axetil can produce preventive analgesic effect, and as thedose increased, analgesia effect enhanced;(2) By comparision, preoperative analgesia groupcan save drugs, it can be considered to use before short surgery; (3) By giving small doses(for the experimental animals, 1mg/kg is a very small dose) ,the weak analgesia of flurbiprofen can be observed,it may be have something to do with the target effect offlurbiprofen.(4)Using high dose (9mg/kg) preoperative , the cumulative pain score is only 6points or so, c-fos-positive neurons are few, so the analgesic effect of flurbiprofen at this doeshas been very perfect. However, if high doses in clinical use,the side effects of flurbiprofenand ceiling effects must be take into account .
