Objective Endotoxaemia is the pathological basis of many kinds of serious infectious diseases, the basic reason that leads to increase infectious disease and many complications. Then, how to control the development of endotoxaemia effectively is one of the keys to reduce the mortality rate of many serious infectious diseases. TCCT, which is originated from "Shang Han Lun" authored by Zhang Zhongjing, is the representative prescription of Tong li gong xia. Recent studies reported that TCCT is widely used in the treatment of all kinds of infectious diseases, apart from its traditional effects. This paper establishes the endotoxaemia by cecal ligation and puncture (CLP), observes the TCCT's therapeutic action on the endotoxaemia, and mortality in endotoxemia of mice, and discusses its mechanisms.Method In this experiment,50 rats are divided into 5 groups at random, those are sham operation group, model group, TCCT group, Radix Et Rhizoma Rhei group and Magnolia officinalis group. Rats are given drugs by intragastric administration 2d before operation, 1h after the last administration, establishes the endotoxaemia by cecal ligation and puncture (CLP), and observes rats'basic symptoms, physical signs, and histomorphology of heart and liver. Establishes a experimental method to determine the content of endotoxin in rats' plasma by the detector BET-72, compares the variation among different groups, and studies the TCCT'function of curing the endotoxaemia. Through detecting the variation of inflammatory cytokines, TNF-α, IL-1βand IL-10 in the rats'serum, discusses the mechanism of TCCT when curing the endotoxaemia.Result Compared with sham operation group, the content of plasma endotoxin is risen significantly in the model group, Radix Et Rhizoma Rhei group and Magnolia officinalis group(P<0.01), so it shows that the model is successfμl, compared with the model group, the content of plasma endotoxin in the test group is decreased obviously(P<0.01), it explains that the test group has a obviously curative effect on the endotoxaemia; compared with the TCCT group, the content of plasma endotoxin in the Radix Et Rhizoma Rhei group and Magnolia officinalis group is larger, which has a significant difference(P<0.05), it states that the treatment effect of TCCT group is better than that of a single Chinese herb; According to the resμlts of the splanchnic tissue before and after having drugs, each group can obviously improve the inflammation and blood stasis of the liver cell, especially the TCCT group, compared with sham operation group, the content of TNF-a and IL-1βare risen obviously(P<0.01), other groups also have an increasing value(P<0.05); compared with the model group, the content of TNF-a and IL-1βare reduced remarkably(P<0.01). compared with sham operation group, the content of IL-10 in the model group and test group are risen obviously(P<0.01); compared with the model group, the content of IL-10 is reduced obviously, which has a significant difference(P<0.01).Conclusion From the experiment results, the TCCT group, Radix Et Rhizoma Rhei and Magnolia officinalis all have antagonistic effect against the endotoxemia, Significantly lower mortality rate of mice, and can improve the visceral injury caused by endotoxin. Among them, the TCCT group is the best one, whose mechanism is relevance to antagonize the inflammatory cytokines such as TNF-α, IL-1βand adjust the ratio of the IL-1β/IL-10.
