| [Objective] According to different resonance response frequency between normal cells and tumor cells by peaking pulse, to choice the best peaking pulse parameter so that tumor cell membrane generate irreversible electrical breakdown induce cell death, and safeguard normal cells as far as possible. To decide the best peaking pulse parameter, compare normal cells and tumor cells after lethal effect, include appearance,cell cycle,invasiveness and initial approach for vivo transplanted tumor model, so that to provide experimental data for lethal effect correlation mechanism,and establish rationale for oncotherapy by peaking pulse.[Method] Experiment object adopt YTMLC that to establish by our own experiment center and health adult lymphocyte (LY),to exert peaking pulse on YTMLC and LY by change different parameter(frequency,time,voltage),to detect YTMLC and LY death rate by MTT and fluorescence staining counting method, determine the best peaking pulse parameter so that to destroy YTMLC and safeguard LY as far as possible. To use the best parameter peaking pulse exert on in vitro YTMLC and LY simulation surroundings, observe YTMLC and LY ultrastructure by transmission electron microscope, detect YTMLC and LY cell cycle by flow cytometer.To detect invasiveness that YTMLC add in invasion cave after action by the best parameter peaking pulse. To use the best parameter peaking pulse exert on vivo transplanted tumor model local and extensive,detect rats survival state and transplanted tumor change,and further understand tissue texture by pathological section.[Results]1,Different parameter peaking pulse exert on YTMLC and LY,detect every sample death rate by MTT and fluorescence staining counting method,the largest difference value between YTMLC and LY death rate when peaking pulse parameter was 120Hz,21kV and 80s,it was the best peaking pulse parameter for the experiment scilicet.2,The best parameter peaking pulse exert on in vitro YTMLC and LY simulation surroundings, according to transmission electron microscope,YTMLC irreversible electrical breakdown effect was apparente and no apparente irreversible electrical breakdown effect on LY;according to flow cytometry,YTMLC to appear S period block,cell proliferation inhibitive and G2/M period cells disappear, however,it was no marked change for LY cell cycle.3,YTMLC invasiveness attenuated markedly by the best parameter peaking pulse.4,To use the best parameter peaking pulse exert on vivo transplanted tumor model local and extensive,the tumor growth restrain obviously(tumor volume decrease,cancer nest necrosis), nevertheless,it was no obviously effect for rats to exist.[Conclusion] The largest difference value between YTMLC and LY death rate when peaking pulse parameter was 120Hz,21kV and 80s,it have clinical meaning obviously.
