| | Effects Of Different Doses Of Puerarin On The Expression Of Bcl-2 And P53 In Rats' Brain Mantle After Cerebral Ischemia/reperfusion |  | Posted on:2010-07-28 | Degree:Master | Type:Thesis |  | Country:China | Candidate:X R Li | Full Text:PDF |  | GTID:2144360302458188 | Subject:Neurology |  | Abstract/Summary: |  PDF Full Text Request |  | Objective:To observe the expression of Bcl-2 and P53 after focal cerebral ischemia/reperfusion in rats brain mantle and to study the effect of different doses of puerarin(Pur) on the expression,for the laboratory basis of the treatment of cerebral infarction and the clinical applications of different doses of puerarin.Methods: 48 SD rats were randomly divided into sham operated(SO) group,cerebral ischemia/Reperfusion (I/R) group,low-dose puerarin(LDP) group,high-dose puerarin(HDP) group. 12 rats were in each group.Rats CI/R models were made by modified method of MCAO.Puerarin intervened.Neurological signs of all rats were scored under Bederson' standard.TTC(triphenyl tetrazolium chloride) staining of rats' brain were done and the average infarct volume were calculated.The number ofapoptotic neurons(terminal-deoxynucleotidy transferase mediated dUTP nick end labeling,TTC staining) and the expression (immunohistochemistry,IHC) of Bcl-2 and P53 in rats' brain mantle were statisticed and compared between each group.Results:1.The neurological action scores of LDP group were lower than that of I/R group(P<0.05).The neurological action scores of HDP group were lower than that of LDP group(P<0.05) and I/R group(P<0.01).2.The average infarct volume of LDP group was lower than that of I/R group(P<0.05).The average infarct volume of HDP was lower than that of LDP group(P<0.01) and I/R group (P<0.01).3.The number of apoptotic neurons of LDP group were lower than that of I/R group(P<0.01).The number of apoptotic neurons of HDP group were lower than that of LDP group(P<0.05) and I/R group(P<0.01).4.The average gray level of Bcl-2-positive cells of LDP group were lower than that of I/R group(P<0.01).The average gray level of HDP group were lower than that of LDP group(P<0.01) and I/R group(P<0.01).5.The average gray level of P53-positive cells of LDP group were higher than that of I/R group (P<0.01).The average gray level of HDP group were higher than that of LDP group (P<0.01) and I/R group(P<0.01).Conclusion:1.Pur can upregulate of Bcl-2 expression and downregulate of P53 expression to protect nerve cells by anti-apoptosis during cerebral ischemia/reperfusion.2.The effect of Pur on expression of Bcl-2 and P53 were dose-dependent.Effect of the high dose Pur was better than that of the low dose. |  | Keywords/Search Tags: | cerebral ischemia/reperfusion, SD rat, Bcl-2, P53, puerarin |  |  PDF Full Text Request |  | Related items |
