Objective: To observe the expression rules of VEGF, Ang-1, Ang-2 after focal cerebral ischemia-reperfusion in rats and to study the effect of puerarin on neurological severity scores, cerebral edema, cerebral infarction, MVD and the expression interventions of puerarin on VEGF, Ang-1, Ang-2 after injected different dose to model rats by different time point. To explore the mechanism of protective effect of Puerarin on cerebral infarct and guide the clinic use time of puerarin.Methods:1. CIR model was established by middle cerebral artery occlusion (MCAO) in rats with nylon thread. The models realized reperfusion after 2 hours ischemia. After anesthesia and citromint heart filling, around 2cm thick brain (the most serious part) samples peri-optic chiasma were selected at 0 hour, 1 hour, 2 hour, 4 hour, 8 hour, 12 hour, 24 hour, 48 hour, 72 hour after reperfusion. Comparing with sham group, observe the expression changes of VEGF, Ang-1 and Ang-2 in different time point samples by immunohistochemistry. Low, peak and descent time points of VEGF were selected as drug administ ration time points.2. Puerarin was selected and divided into low, middle and high dose groups (27mg.kg-1, 54mg.kg-1 and 108mg.kg-1). Puerarin was injected by abdominal cavity at 0 hour, 24 hour and 48 hour three time points and solvent group injected 50% Propanediol once per day for one week.
