Objective Clinical gastric cancer are common malignant tumor,which is a kind of cell proliferaton and apoptosis are abnormal happened diseases.Phosphatase and tension homologue deleted on chromosome 10(PTEN) as a tumor suppressor gene, cyelooxygenase-2(COX-2) and Survivin to inhibit tumor apoptosis gene.by detecting them in the normal gastic mucosa and gastric carcinoma tissue to study the occurrence and development of gastric cancer in the mechanism and clinical significance of mutual relations.Research subjects and methods The collection of the Jinan Military Region General Hospital from May 2006 to April 2008 surgical resection or endoscopic biopsy specimens of a total of 95 cases,15 cases of normal group,their aged 47-75 years old,average 51.3. 80 cases of gastric cancer group,patients aged 52-78 years old,average 69.2.According to the degree of tumor differentiation into well-differentiated groups,differentiated and poorly differentiated group.Gastric cancer staging adopted in 1997 UICC 5th edition gastric TNM staging.Immunohistochemical S-P method for testing.Kit operate according to instructions.Positive results to determine:number of positive cells accounted for by the proportion of total cells to determine,select the 10 high-power microscope field of vision, counting the number of positive cells,positive cells<10%(-);10-25%(+);25-50%(++);>50%(+++)。Results 15 cases of normal gastric mucosa were observed strong expression of PTEN protein,and 80 cases of gastric cancer of positive expression rate of 47.5%(38/80), significantly lower than that in normal gastric mucosa(P<0.05),PTEN-positive rate atⅠ-Ⅱphase and without lymph node metastasis in gastric cancer was significantly higher than that have lymph node metastasis andⅢ-Ⅳgastric cancer,the difference was significant(P<0.05),with the degree of tumor differentiation and depth of invasion has nothing to do.COX-2 in gastric cancer positive rate of 67.5%(54/80),no expression in normal gastric mucosa(P<0.05);at have lymph node metastasis and TNM stagesⅢ-Ⅳgastric carcinoma period,the positive was significantly higher than that without lymph node metastasis and TNM stagesⅠ-Ⅱperiod of gastric cancer tissues(P<0.05);but the depth of invasion and differentiation.Survivin in gastric cancer positive rate of 70.0% (56/80),with the degree of increased malignant tumor positive rate was increasing trend in the poorly differentiated group was significantly higher than in well-differentiated group, TNM stagesⅢ-Ⅳperiod have lymph node metastasis was significantly higher thanⅠ-Ⅱperiod and without lymph node metastasis group,the differences were statistically significant(P<0.05).PTEN positive expression cases,COX-2 and Survivin positive rate was significantly lower than the expression of PTEN-negative group,the difference was significant(P<0.05).Conclusion PTEN with the COX-2 and Survivin expression was negatively correlated(P<0.05),and COX-2 and Survivin expression was positively correlated(P<0.05),PTEN deletion and decreased expression and COX-2,Survivin over-expression between there may be mutual synergies involved in the occurrence and development of gastric cancer, metastasis and infiltration of the three biological factors of the Joint Detection,not only helps to determine the degree of malignancy of gastric cancer,and contribute to tumor pathogenesis and provide a theoretical basis...
