| Objective: To observe the effect of yizhi jiannao granule concentration fluid(YCF) on the expression of Neuroglobin and Bcl-2 mRNA in brain tissue of amyloid-β25-35 brain injection induced experimental AD model rats in vivo,this work discusses partially the mechanism of traditional Chinese medicine YCF with respect to its effectiveness to treat Alzheimer's disease.Methods: 40 SD rats were enrolled in the study, and they were randomly divided into four group: blank controls group(n=10), sham controls group(n=10), model group(n=10) and YCF group(n=10). A rat model of AD was induced by amyloid -β25-35 sterotaxis injection into the bilateral Hippocampus. They were cured for twenty-eight days, then their ability of learning and memory was detected by Morris water maze experiment and the expression of Ngb and Bcl - 2 mRNA in brain tissue was assayed by real-time quantitative reverse transcription - PCR.Results: 1. Result of praxiology experiment showed that the learning and memorizing performance decreased in model group ,and then the escape latency time was significantly shortened, the time of swim in the platform quadrant significantly increased in the YCF group (p<0.05) . 2.The expression of Ngb mRNA of hippocampus tissue of rats in model group than that in sham-operation group (P<0.05 ) ,and that in YCF group was signifieantly higher than that in model group (P<0.01) .3.The expression of Bcl-2 mRNA of hippocampus tissue of rats in model group than that in sham-operation group (P<0.05) ,and that in YCF group was signifieantly higher than that in model group (P<0.01) .Conclusion: 1. The results of the study show that the model which induced by amyloid -β25-35 sterotaxis injection into the bilateral Hippocampus is reliable animal model of Alzheimer's disease . 2. The learning and memorizing ability of the model rats were decreased obviously, Yizhi jiannao granule can improve the learning and memorizing ability of the model rats(P<0.05). 3. Yizhi jiannao granule may up - regulate expression of Ngb mRNA Bcl-2 mRNA, which could be the remedial mechanisms of Yizhi jiannao granule in the treatment of Alzheimer'disease through neurovascular protection. |
