Objective:To study and explore the neuroprotective effects and the further mechanism of aloe polysaccharide on cerebral ischemia.Methods:KM mice were randomly divided into six groups:high-dose group, middle-dose group and low-dose group of AP.positive control group:Nim,GLT.models control group.Every mice was fed with drugs for seven days.After giving drugs for 1 hour, To use different methods to record BT,CT and DGT,Wistar rats were randomly divided into seven groups:high-dose group,middle-dose group and low-dose group of AP. positive control group:Nim,GLT.models control group:sodium chloride.sham group. Every rat was fed with drugs for seven days.After giving drugs for half an hour,In order to observe the neuroprotective effect of aloe polysaccharide on cerebral ischemia,the rats with incomplete cerebral ischemia were induced with fishing lines in the middle cerebral artery(MCA) and then were executed at 24h after cerebral ischemia.It was observed the effect of aloe polysaccharide on the changes of ethology and pathomorphology due to nerve injury of rats with MCAO.HE staining was used to observe the changes of histomorphology.It was detected the effects of aloe polysaccharide on protein expression of heat shock protein70(HSP70),protein kinaseB(PKB) with immunohistochemical method.Results:Middle-dose group of AP can prolong BT,CI\DGT of KM mice;Aloe polysaccharide could make obviously pathomor pholo- gy of cerebral ischemia better, improve change of ethology,increase the expression of HSP70,PKB protein in cerebral cortex.Conclusion:Middle dose group of AP can promoting blood flow,anticoagulated blood and prolong DGT.the mechanism is related to cutting down the oxygen consumption.Aloe polysaccharide had protecting effect on cerebral ischemia and the mechanism of aloe polysaccharide against cerebral ischemia is related to affecting the related expression of gene and protein interrelated with cell apoptosis.
