Objective: It was studied and explored further the mechanism of treatment that the neuroprotective effect of phenolic alkaloids from Menispermum dauricum (PAMD) on cerebral ischemia in rats.Methods: In order to test the neuroprotective effect of PAMD treating cerebral ischemia, the rats with incomplete cerebral ischemia were induced with fishing line in the middle cerebral artery occlusion (MCAO) . It was detected the effect of PAMD on protein expression of intercellular adhesion molecule-1 (ICAM-1) , Cyclooxygenase-2 (COX-2) , Nuclear Factor Kappa B p65 (NF-κB p65) and cysteiny aspartate specific protease-3 (Caspase-3) with immunohistochemical process at 24h, 48h and 72h after cerebral ischemia, and the gene expression of Caspase-3mRNA with reverse transcription polymerase chain reaction (RT-PCR) and rate of cell apoptosis with flow cytometry at 72h after cerebral ischemia, and it was also observed the effect of PAMD on the change of ethology and pathomorphology due to nerve injury of rats with MCAO before and after treatment.Results: PAMD could make obviously pathomorphology of cerebral ischemia better, improve change of ethology, inhibit expression ICAM-1, COX-2, NF-κB p65, Caspase-3 and Caspase-3mRNA, and decrease the rate of cell apoptosis of ischemic cerebral tissue. The best therapeutic effectiveness was observed in high dose group of PAMD, and the better therapeutic effectiveness was also got with longer treatment course.Conclusion: PAMD had certain both time-effect and dose-effect relationship in protecting effect on cerebral ischemia. The mechanism of PAMD against cerebral ischemia is related to anti-inflammatory action, inhibiting cell apoptosis and affecting the related gene expression interrelated with cell apoptosis.
