Objective:To study the relation on syndrome differentiation in Hyperplasia of Mammary Gland and the expression of P53,P16,and research the pathogenesis of HMG.We hope to provide theoretical basis for objective,normative standards of diagnosis for HMG.Methods:Fourty-one cases of HMG were selected randomly and divided into three groups according to relevant criteria adopted by surgical branch committee of Chinese Medical conference of TCM in 2002(Stagnation Liver—qi type,Turbid Phlegm and Blood stasis type,Disorder Thoroughfare vessel and conception vessel type).Main syndromes and the different symptoms were observed in three groups;level of E2,LH,P,FSH,TT and PRL were determined in order to observe the changes of sex hormones in different groups and analyze the relationship of the changes with syndrome differentiation;Western blot analysis were performed to determine the expression of P53,P16 in proliferation tissues and normal breast tissues.Results:1.There was obviously difference of the shape,texture,scope among those three groups, but there wasn't obviously difference in the site morbidity.2.Compared with normal group,the level of LH and E2 were significantly decreased in the patient of HMG,but the level of P,FSH,TT,PRL did not significantly change,the level of E2 and P in Stagnation Liver—qi type were higer than that in Disorder Thoroughfare vessel and conception vessel type,there wasn't obviously difference between Stagnation Liver—qi type and Turbid Phlegm and Blood stasis type,Turbid Phlegm and Blood stasis type and Disorder Thoroughfare vessel and conception vessel type.The level of LH in Turbid Phlegm and Blood stasis type was higher than that in Disorder Thoroughfare vessel and conception vessel type,there wasn't obviously difference between Stagnation Liver—qi type and other two groups,the level of PRL in Stagnation Liver—qi type and Disorder Thoroughfare vessel and conception vessel type were both lower than that in Turbid Phlegm and Blood stasis type,there wasn't obviously difference between Stagnation Liver—qi type and Disorder Thoroughfare vessel and conception vessel type.There wasn't obviously difference in three groups of the level of FSH and TT.3.The expression of P53 in proliferation tissues was higher than that in normal breast tissues,and the expression of P53 in Turbid Phlegm and Blood stasis type was lower than that in Stagnation Liver—qi type and Disorder Thoroughfare vessel and conception vessel type,but there wasn't obviously difference between Stagnation Liver—qi type and Disorder Thoroughfare vessel and conception vessel type4.The expression of P16 in proliferation tissues was lower than that in normal breast tissues,and the expression of P16 in Disorder Thoroughfare vessel and conception vessel type was lower than that in Stagnation Liver—qi type and Turbid Phlegm and Blood stasis type,but there wasn't obviously difference between Stagnation Liver—qi type and Turbid Phlegm and Blood stasis typeConclusions:1.There was obviously difference of the shape,texture,scope among those three groups which shows that can would a objective indicator for syndrome differentiation.2.The patient of HMG manifests endorine dysfunction,and patients with different TCM syndrome were different in secretion of sexual hormones,which shows sexual hormones would affect the occurrence and development of HMG to some extent.The changes of sexual hormones would be use as objetctive indexes of syndrome differentiation and basis of clinical medical application.3.The abnormal expressions of P53,P16 show that would be have a significant impact on the course of the occurrence and development of HMG.,and the different expression in three groups show that would be use as objetctive indexes of syndrome differentiation from the protein level. |
