Effect Of Chang Yu Ning On IL-8 Of Ulcerative Colitis In Rats And Related Clinical Research Of This Disease

Objective:By observing the therapy effect of Chang-yu-ning granule which can heat-clearing and detoxicating,invigorate the spleen and deprive the evil wetness on ulcerative colitis,to discuss the basic pathology mechanism of ulcerative colitis and analyse the potential mechanism of action of Chang-yu-ning granule.To provide a theoretical principle for clinic.Methods:1.Experimental research150 Wistar rats are randomly divided into five groups.There are blank control group,model control group,western medicine contol group,high dose and low dose groups of Chinese medicine.To prepare the rat model of ulcerative colitis by using 2,4-dinitrochlorobenzene and acetic acid and feeding high fat diet.Rats in every group are giving intragastric administration for four weeks.Western medcine control group took salicylazosulfapyridine,while high dose and low dose groups of Chinese medice take chang-yu-ning granule,blank control group and model contral group take distilled water.To survey Interleukin-8 levels of rats colon tissue by enzyme linked immunosorbent assay.To observe rats intestinal mucosa's change by native eyes.To grade inflammatory activity index and the general form of injury.2.Clinical research41 cases of ulcerative colitis patients are radomly diveded into test group and control group,We applied the Chang-yu-ning granule combined with Salicylazosulfapyridine Tablets to treat 21 cases in test group,while 20 cases in control group are taken the Salicylazosulfapyridine Tablets only.30 days is one threapy process,and they should take two processes in succession. According to curative effect determinate standard to compare the thereapy effect of two groups. Result:1.Empirical study resultIL-8 result indicates that each test group IL-8 level has obviously declined,compared wih model control group it has statistical significance(P＜0.01).In the western medcine contol group,high dose and low dose groups of Chinese medice,the grades of inflammatory activity index and the general form of injury have decreased,comparede with modle contol group has statistical significance(P＜0.01),while difference of each test group is nonsense.The pathomorphology result indicates that compared with model contol group the colon mucosal ulcer,mucosal tissue edema,hyperaemia and inflammation cell in each test group have alleviated.2.Clinical research resultAfter having thereapy each group cases' traditional Chinese medicine symptom overall integral has declined than their thereapy ago.It is statistical significance(P＜0.01).And the symptom overall integral in test group has superiority to the control group.The main symptoms of the efficacy and points indicate that 3 symptoms in 8 of test group are better than control group(P＜0.05).The other 5 symptoms in test group are also better than control group(P＜0.01).Conclusion:Chang-yu-ning granule has definite curative function on both animal model and patients of ulcerative colitis.It's action mechanism is potentially to lower colon tissue IL-8 level,to improve inflamation action index,to alleviate colon mucosa ulcer,to reduce inflammatory cell infiltrate. So it has the function of curing ulcerative colitis.
