Mulberry fruit is by-product of silkworm rearing.As a traditional Chinese herbal medicine,more and more people are paying attention to its pharmacological activities and empoldering of mulberry fruit.It can nourish yin and supplement blood,clear heat and promot fluid production,prolong life,etc.It can treatment insomnia,constipation,tinnitus,white beard and hair as early as,too. Polysaccharide is one of the most effective medicinal activity elements of mulberry fruit.Based on research,we discovered that mulberry fruit has antioxidant activity, hypoglycemic activity,anti-tumor activity,anti-inflammatory activity,etc.In addition,as a natural plant,mulberry fruit show no negative effects to human health. Therefore,the utilization of mulberry fruit shows a broad market demand.In this study,we optimized the extraction parameters of FMAP.In the processes of extraction,we set four factors as extracting temperature,extracting time, solid-liquid ratio and extracting times,and by single-factor experiments to ascertain the conditions of orthogonal test.The optimum extracting conditions was temperature 80℃,solid-liquid ratio 1:30,2 times,4 hour per time.Under this condition,the extracting ratio was 5.71%.The DEAE-cellulose52 column chromatography was used to separate FMAP.The largest eluting peak by using solution of 0.2M NaCl was obtained,concentrated, freeze-dried,named as FMAP-2.With High-Performance Gel Chromatography (HPGPC),the FMAP-2 was demonstrated as a heteropolysaccharide,and the average molecular weight was 3.0×105Da.With Fourier Transform Infrared Spectroscopy(FTIR) analysis,the FMAP-2 showed absorption peaks at 3411,2922, 1623,1437 and 1099,which are identical to the functional groups as -OH,-CH2-, -CHO,-COOH and C-O-H,respectively.The GC and GC-MS analyses show that FMAP-2 from mulberry fruit was composed of glucose(34.8%),talose(25.47%), arabinose(22.78%),galactose(8.60%),mannose(4.49%) and fructose(3.87%). We used hyperglycemia mice which were alloxan-induced by the tail vein injection to study hypoglycemic activity of FMAP-2.FMAP-2 administered orally for 4 weeks,we found the blood glucose levels of mice from 23.8mmol/L to 15.7mmol/L,and the blood glucose levels of normal mice never show any significant change.
