As people life rhythm speeded up and working pressure increased, there are more and more patients with diabetes, which present a younger trend. Now diabetes has become one of the major diseases that threaten human. Diabetes is a disease that needs a lifelong treatment, and difficult to be cured. Oral administration of synthetic drugs was mostly used to treat diabetes in addition to insulin injections, which could not be stopped and would bring harm to persons` liver. It's very meaningful that to seek cure drugs for diabetes from the traditional Chinese medicine, especially plant resources. The modern pharmacological experiments indicated that the polysaccharide, flavonoids and alkaloids from mulberry leaves could obviously decrease the contents of blood glucose of experimental diabetic mice. The polypeptide from balsam pear has obviously lowering blood glucose activity, called"the plant insulin". It was studied that response time of the traditional Chinese medicine ingredient was longer but with slower effect. The hypoglycemic effect of the union use of Chinese herbs and western drugs was studied in the experiment after the hypoglycemic effect of the polysaccharide, flavonoids and alkaloids from mulberry leaves was validated.Firstly, the polysaccharide, flavonoids and alkaloids from mulberry leaves and the polypeptide in balsam pear were extracted separately. And the extraction technologies of conditions of mulberry were optimized. The optimal extraction conditions were gotten. All the pure samples for the next pharmacology investigation step were prepared after the corresponding examination.Then, the hypoglycemic effect of the polysaccharide, flavonoids, alkaloids from mulberry leaves and the polypeptide in balsam pear was observed and compared. And the blood glucose was determined. Influences of the blood fat, glycogen, blood glucose and glucose tolerance were overall evaluated simultaneously. Effects of the polysaccharide, flavonoids and alkaloids in mulberry leaves and the polypeptide in balsam pear on the blood glucose, blood fat and glucose tolerance of streptozocin diabetic rats were studied in this study. Results showed that the contents of blood glucose could be obviously decreased and the glucose tolerance could be evidently increased by the polysaccharide, flavonoids and alkaloids in mulberry leaves and the polypeptide in balsam pear, which can against the dyslipidemia associated with the diabetes. The glucose decline rates were 14.42%,27.33%,37.55% and 30.71% respectively. It was demonstrated by all above results that the mulberry polysaccharide, flavonoids and alkaloids could prevent and treat diabetes. And the alkaloid in the mulberry leaves has obvious hypoglycemic effect.Influence of the effective ingredients in mulberry leaves on blood glucose, glycogen and glucose tolerance of diabetic rats when combinated with metformin was investigated based on this research. The combined medication was administered once a day for 12 days to observe their effects on blood glucose, weight and glycogen in diabetic rats. Diabetic rats were treated using different combined medicines and metformin was administered at the dose of 100 mg/kg to another group as the positive reference substance. The results indicated that the polysaccharide, flavonoids and alkaloids in mulberry leaves could decrease the contents of blood glucose in diabetic mice to a certain extent when combinated with metformin, And the alkaloids ingredient have obvious hypoglycemic effect when combinated with metformin. And the glucose decline rate was 22.86%, which has improved 16.53% compared with the groups administered the metformin only. Theory basis and the experiment supports were provided for the further development of mulberry to becoming the healthcare products and functional food which could control the blood glucose and the complications of the diabetes.
