Background ApoE is a plasma protein that play an important role in the metabolism of cholesterol an other serum lipids.It is an important determinant of risk for the development of atherosclerosis.in addition to this effect,it is proposed that one of the major functions of ApoE is mediate neuronal repair,remodeling and protection.Although theε4 allele has been found to be consistently associated with risk of AD,the relationship between ApoE allele and aerebrovascular disease is controversial.Objective To analyze association between the polymorphisms of apolipoprotein E(ApoE) and risk for ischemic cerebrovascular disease(ICVD) in Bai populations from Yunnan Province,and to compare with each group's difference in alleles in in serum lipid levels.Methods We used PCR-RFLP method to detect ApoE genotypes of 40 patients with ischemic cerebrovascular diseases(male17cases,23cases of wome -n),and contrasting with that of age and sex-matched normal group(male 25 cases, female 18 cases).And the concentration of apoE in patients with ACI and in control subjects was determined.Meanwhile we detected their plasma lipid levels and hemorheological characteristics.At last,we used gene sequencing to verify the ApoE gene polyrnorphism with 30 cases(ICVD patients with 15 cases,15 cases of normal controls).Results(1) In Bai normal contrasting group,the TG value ofε2 andε4 were 0.97±0.18mmol/L,1.46±0.12mmol/L respectively,the HDL value ofε2 andε4 were 1.27±0.21 mmol/L,1.13±0.27mmol/L respectively.In Bai ICVD group,the TG value ofε2 andε4 were 1.35±0.68mmol/L,TG2.04±0.65mmol/L respectively,the HDL value ofε2 andε4 were 1.47±0.27 mmol/L,1.09±0.30mmol/L respectively.(2)In Bai ICVD group,the genotype frequencies of E3/3,E2/3,E3/4 were 0.600,0.150 and 0.225 respectively.In Bai control group,the genotype frequencies of E3/3,E2/3, E3/4 were 0.744,0.163 and 0.047 respectively.In Bai ICVD group,the genotype frequencies ofε2,ε3 andε4 were 0.088,0.787 and 0.125 respectively.In Bai control group,the genotype frequencies ofε2,ε3 andε4 were 0.071,0.744 and 0.185 respectively.There were no differences betweenε4 andε3.(3)Different cerebral infarction patient carry differentε4 allele frequency.they were cardiogenic cerebral embolism(50.0%),atherosclerotic thrombotic cerebral infarction(74.1%),lacunar infarction(11.1%).and there was significant difference between atherosclerotic thrombotic cerebral infarction group and lacunar infarction group(P<0.05).(4)In male ICVD group,the genotype frequency of E3/4 was 0.294.In female ICVD group,the genotype frequency of E3/4 was 0.174.There were no differences between male ICVD group and female ICVD group(x~2= 3.369,P=0.066 ).In male control group,the genotype frequency of E3/4 was 0.040.In female control group, the genotype frequency of E3/4 was 0.055.There were no differences between male control group and female control group(x~2=0.222,P=0.637).Conclusions(1) Our data show an association between APOE gene polymorphism and the onset of ICVD and support the possibility that epsilon4 allele is a susceptibility locus for the risk of ICVD.ApoE plays a key role in the metabolism of cholesterol and other serum lipids.The effect of ApoE on ICVD may be mediated through dyslipidemia and atherogenesis through its effect on cholesterol and triglyceride metabolism.(2)ε4 allele is correlated positively with TC and LDL-C and negatively with HDL-C,oppositeε2 allele.(3)ε4 allele is correlated with athero-sclerosis -related thrombosis,and irrelevant with lacunar infarction.(4) There was no difference in gender between Bai ICVD group and Bai control group.(5) We used both PCR-RFLP methods and gene sequencing method to detect ApoE genotype,the results are simillar;but gene sequencing is more accurate,rapid,reliable,and missed the low rate of repeat good and so on.
