Study On The Effect Of The Treatment Of Invigorating Spleen And Promoting Blood Regulating Qi On Pancreat Tissue-Apoptosis Of The Chronic Pancreatitis Fibrosis Rats

Objective:Based on Preliminary research,our discussion group further observed the influence of the treatment principle of "invigorating spleen and promoting blood by regulating qi" in Pancreatic tissues in experimental animals,we establish the rat model of chronic Panereatitis(CP).This researeh Tried to study and analyse the expression of Bcl-2,Bax and NO when CP occurred,to penetrate into discussion about tMs principle's anti-fibrous mechanism.Method:Sixty SD rates were divided into 5 groups randomly:normal control group,Pathology model group,Qing yi li dan treatment group,high dose group of "YI TAI Compounded Medication" and low dose group of "YI TAI Compounded Medication".Each group had 12 rats.To establish the chronic pancreatitis rats model with 13.3%L-arginine with the method of celiac injection,10 days after,we gave them "YI TAI Compounded Medication".High dose group of "YI TAI Compounded Medication" were perufsed stomach with 11.25/kg everyday,low dose group of "YI TAI Compounded Medication" were perufsed stomach with 5.63g/kg everyday,and the samples of pancreas were collected after 2months for evaluating.Utilizing the techniques,such as immuno- histochemistry,optical microscope and hybridization in situ,the paper investigated the effect of the regulating factors of texture fibrous Bcl-2,Bax and NO's contents of serum in CP rats; meanwhile,observes the intervening effect of "YI TAI Compounded Medication".This paper studied the anti-fibrous mechanism from tissue form and molecular level.Result:1.Our research injected L-Arg into abdomen to make Spleendeficiency rat model,According with spleen deficiency syndromeand Chronic Pancreatitis from symposium,physical sign and tissueform,so this rat model is sucessful.2.The treatment principle of "invigorating spleen and promoting blood by regulating qi" could recover most of the pancreas acinar, reduce fiber constituents in pancreas lobular,accelerate absorption of inflammatory cells,increase body's dissolved ability;3.The treatment principle of "invigorating spleen and promoting blood by regulating qi" could restrain the expression of Bcl-2/ Bax,elevating the Apoptotic index,so as to promoting apoptosis,and deposition of ECM;4.The treatment principle of "invigorating spleen and promoting blood by regulating qi" could increase the expression of NO to dissolved fiber to accelerate the degradation of ECM.Conclusion:The treatment principle of "invigorating spleen and promoting blood by regulating qi" is an effective method of preventing and treating CP, and YI TAI Compounded Medication has a higher clinical value.Its action implements through influencing apoptosis,regulating the synthesis and degradation of ECM,so as to protected pancreatic cells and restrain the fibrosis of pancreas.
