Objective: To investigate the effect of Curcumin on the expression of Pleiotrophin(PTN) in cervical cancer Caski cells and the biological behaviour.Methods: The cervical cancer Caski cells were cultured in vitro ; The immunocytochemistry was used to assay the distinctive expression of PTN in Caski cells which were treated by Curcumin or not ; The Western blotting was used to assay the effects of Curcumin with different concentrations on the protein levels of PTN , and the end levels of Curcumin were 10μg/ml,20μg/ml,40μg/ml ; MTT was used to assay the proliferation of Caski cells which were treated with different concentrations of Curcumin ; The transwell boyden chamber invasive test was used to assay the variation of the ability of Caski cells treated with different concentrations of Curcumin .Results:①The immunocytochemistry test showed that the expression of PTN was hadro-positive in Caski cells; The buffy grains aligned in cytolymph and cell nucleus; Contrast with the control team , the expression of PTN was significant down-regulated after treated with Curcumin.②The Western blotting showed that the Curcumin could down-regulate the expression of PTN (P<0.05), and this was a does-dependent manner.③The proliferation of Caski cells were inhibited by Curcumin with a does-dependent manner , and the inhibition ratios were 39.5%,54.4%,60.4%(P<0.05).④The boyden chamber invasive test showed that with the concentrations of Curcumin increased, the number of Caski cells which transwell was decreased obviously.Conculsions: Curcumin could inhibit the proliferation,the invasion and aversion ability and the PTN expression of Caski cells significantly, and those changes were does-dependent (p<0.05).This maybe could provide a new thought for adjunctive therapy of the cervical cancer.
