Hyperlipidemia Fatty Liver is one of the most common kinds of Nonalcoholic liver disease (NAFLD), which excessive fat storing in liver and hepatic cellular degeneration without over ingestion of alcohol. Great progress has been made in the study of fatty liver with integrated medicine of traditional Chinese and western in aspects of diagnosis, treatment and experimental study, etc. According to the pathological mechanism of traditional Chinese medicine (TCM), fatty liver is characterized by deficiency in nature and repletion in appearance. In recent years, the incidence of fatty liver is increasing quickly. In some patients, it can transform into liver cirrhosis, liver cancer or some heart disease. It becomes a serious harmful disease of human being. Many traditional Chinese drugs have the function of strengthening spleen, regulating liver and removing stasis and phlegm, and it has been proved significantly effective in treating hyperlipemia fatty liver through years of clinical observation. NXKKFY is developed from the tradition famous prescription of BYHWT. It is a utility prescription of treatment on hyperlipemia fatty liver due to deficiency of QI in our department (The department of traditional Chinese medicine in JI LIN university. ) We built animal model of hyperlipemia fatty liver and observe the effect on oxygen free radical and liver lipid metabolism. We study the mechanism of NXKKFY and want to develop and utilize it.Methods:80 healthy male SD rats were divided into 6 groups at random, namely, normal control group, model control group(negative control group), Dong-Bao-Gan-Tai control group (positive control group), low-dose group of NXKKFY, midst-dose group of NXKKFY and high dose group of NXKKFY. Except for the ordinary forage in normal control group, the hyperlipemia fatty liver models were built by feed with high-fat forage to the rest of groups. After 8 weeks, hyperlipemia fatty liver models were succeeded. The rats were treated by NXKKFY or Dong-Bao-Gan-Tai by intragastric administration for 4 weeks, rats were killed to observe and analyze the effect of LPL, HL, FFA, MDA and SOD in blood serum and liver homogenate. Pathological sections were dyed to observe the histopathological changes.Results:1. The changes of LPL and HLCompared with normal controlled group, the level of LPL and HL in blood serum and liver homogenate decreased very insignificant (P < 0.01), midst-dose group and high dose of NXKKFY control groups were decreased markedly compared with model group(P<0.01),but difference with normal group(P<0.05). NXKKFY control groups were decreased without statistical difference(P>0.05)compared with Dong-Bao-Gan-Tai control group.2. The changes of FFACompared with normal controlled group, the level of FFA in blood serum increased very insignificant (P<0.01), midst-dose group and high dose of NXKKFY control groups were increased markedly compared with model group(P<0.01),but difference with normal group(P<0.05). Dong-Bao-Gan-Tai control group were decreased without statistical difference(P>0.05)compared with NXKKFY control groups.3. The changes of MDA and SODCompared with normal controlled group, the level of SOD and MDA in liver homogenate and blood serum changed very insignificant. While the level of MDA increased significantly and the level of SOD decreased significantly in negative control group(P<0.01). The low-dose, midst-dose and high dose group of NXKKFY all can increase the level of SOD and decrease the level of MDA at the same time, which are very significant difference compared with negative control group (P<0.01).The midst dose group of NXKKFY have better capability to increase the level of SOD and the high dose group decrease the level of MDA than other dose groups of NXKKFY, but have no significant differences of statistics(P>0.05). All dose group of NXKKFY have no significant differences of statistics by compared with Dong-Bao-Gan-Tai group(P>0.05).4. Liver histological featuresSevere hepatocellular steatosis and inflammation were found in model group compared with normal group, NXKKFY control groups and Dong-Bao-Gan-Tai group were shown improvement. Compared with model group, midst-dose group and high dose of NXKKFY control groups were decreased markedly(P>0.01), Dong-Bao-Gan-Tai control group were decreased without statistical difference(P > 0.05)compared with NXKKFY control groups. Conclusion:NXKKFY can increase the level of LPL and HL in liver homogenate and blood serum, and it can also increase the level of SOD and decrease the level of MDA. At the same time, NXKKFY also can forbid the excessive increase of FFA. As a result, NXKKFY has an effective action on hyperlipemia fatty liver.
